Preparation of cytoskeletal markers and organization of the cytoskeleton during symbiotic interactions of alfalfa (Medicago sativa L.)

Abstract:

Cytoskelet se v rostlinné buňce účastní řady esenciálních procesů, jako je dělení a diferenciace buněk, transport, intracelulární organizace a signalizace. V důsledků vnějších stimulů dochází ke změnám uspořádání jeho dvou hlavních komponent, mikrotubulů a aktinových filament. K reorganizaci cytoskeletu dochází i během symbiotických procesů, kdy dochází ke změně tvaru kořenových vlásků a stimulaci buněčného dělení v kortexu kořene. Cytoskelet také napomáhá při růstu infekčního vlákna a tvorbě symbiotických hlízek, ve kterých dochází k fixaci vzdušného dusíku bakteriemi. Medicago sativa (vojtěška setá) je rostlina z čeledi bobovitých, která se vyznačuje symbiózou s půdními bakteriemi rodu Ensifer se schopností fixovat vzdušný dusík. Symbiotickou fixací je do zemědělských systémů ročně dodáváno několik desítek tun dusíku, čímž se snižuje potřeba aplikace dusíkatých hnojiv. Díky vysoké nutriční hodnotě vojtěška nabyla nezastupitelné místo jako krmivo pro hospodářská zvířata, což z ní činí zemědělsky velmi významnou plodinu. Cílem předkládané bakalářské práce byla příprava markerů tubulinového cytoskeletu vojtěšky metodou klonování Golden Gate. Připravené konstrukty byly transformované do bakterií Agrobacterium tumefaciens pro následující tranzientní transformaci listů Nicotiana benthamiana s účelem ověřit funkčnost markerů. Dalším cílem bylo ověřit funkčnost komerčně dostupného markeru aktinového cytoskeletu. V rámci následující práce budou markery stabilně transformovány do vojtěšky pro mikroskopické studium cytoskeletu během symbiotických interakcí s rhizobiemi.

Abstract:

The cytoskeleton in plant cells participates in many essential processes, such as cell division and differentiation, transport, intracellular organisation, and signalisation. As a result of external stimuli, the arrangement of its two main components, microtubules and actin filaments, changes. Cytoskeleton reorganisation also occurs during symbiotic processes, when the shape of root hairs changes and cell division is stimulated in the root cortex. The cytoskeleton also assists in the growth of infection thread and the formation of symbiotic nodules, where bacteria fix the atmospheric nitrogen. Medicago sativa (alfalfa) is a plant of the Fabaceae family characterised by symbiosis with soil bacteria of the genus Ensifer with the ability to fix atmospheric nitrogen. Symbiotic fixation supplies tons of nitrogen to agricultural systems every year, reducing the need to apply nitrogen fertilisers. Thanks to the high nutritional value, alfalfa gained an irreplaceable place as livestock feed, making alfalfa a significant agricultural crop. The presented bachelor thesis aimed to prepare tubulin cytoskeleton markers of alfalfa by the Golden Gate cloning. Prepared constructs were transformed into Agrobacterium tumefaciens for subsequent transient transformation of Nicotiana benthamiana leaves to verify the functionality of the markers. Another aim was to verify the functionality of a commercially available actin cytoskeleton marker. As a part of the following work, the markers will be stably transformed into alfalfa for microscopic study of the cytoskeleton during symbiotic interactions with rhizobia.
