Telomeric repeats and their interactome

Abstract:

Telomerické repetice se vyskytují nejen na fyzických koncích eukaryotických chromozomů – telomerách, ale také uvnitř těchto chromozomů, kde se nazývají intersticiálními telomerickými repeticemi. V Arabidopsis thaliana bylo popsáno několik rodin proteinů asociovaných s telomerickými sekvencemi. Přesná asociace těchto proteinů s koncovými nebo intersticiálně lokalizovanými telomerickými repeticemi, a jejich funkce (zvláště u pylu), není doposud zcela jasná. Z vybraných proteinových rodin, které by se mohly vázat na krátké intersticiálně lokalizované telomerické sekvence, byly vybrány některé kandidátní proteiny. V teoretické části jsem shrnula základní informace o telomerách, intersticiálně lokalizovaných telomerických repeticích, proteinech asociovaných s telomerickými repeticemi, proteinových komplexech, jež chrání telomery před zkracováním nebo mají vliv na regulaci genové transkripce a vývoj pylového zrna. V praktické části bakalářské práce jsem se zabývala klonováním genů kódujících tyto kandidátní proteiny.

Abstract:

Telomeric repeats occur not only at the physical ends of eukaryotic chromosomes – telomeres, but also inside these chromosomes, where they are called interstitial telomeric repeats. Several protein families associated with the telomeric sequences have been described in Arabidopsis thaliana. The exact association of these proteins with terminal or interstitially localized telomeric sequences and their function (especially in pollen) is not entirely clear yet. Several candidate proteins were chosen from selected protein families that could bind to short interstitially localized telomeric sequences. In the theoretical part I summarized basic information about telomeres, interstitially localized telomeric repeats, proteins associated with telomeric repeats, protein complexes that protect telomeres from truncation or affect the regulation of gene transcription and pollen grain development. In the practical part of the bachelor thesis, I focused on the cloning of genes encoding these candidate proteins.
