Zavedení a optimalizace buněčné linie RLT2 pro výzkum biochemických mechanismů toxicity

Implementation of the cell line RTL2 for studies of biochemical toxicity

Abstract:

In vitro testy jsou vhodné pro zkoumání biologické aktivity látek v různých složkách prostředí. Nejčastěji se využívá testů s reportérovými geny. Výhodou je jejich dobrá citlivost i reprodukovatelnost výsledků, často i jednoduchost testování. Sledovala jsem aktivitu receptoru pro planární aromatické látky (AhR), který hraje hlavní roli ve zprostředkování toxicity dioxinového typu. Následkem vazby na Ah receptor dochází v buňkách k biochemickým změnám, které vedou k regulaci genové transkripce. V experimentech jsem použila rekombinantní buněčnou linii RLT2.0 z hepatokarcinomu pstruha duhového, která je stabilně transfekovaná plazmidem pGudLuc1. Tento plazmid obsahuje luciferázový gen ze světlušky. Po expozici chemickou látkou, v tomto případě 2,3,7,8-tetrachlordibenzo-p-dioxinem (TCDD), dochází k indukci luciferázy, která v přítomnosti svého substrátu vytváří luminiscenční signál, který je úměrný odpovědi buňky. Toxicitu dioxinového typu vykazují například polychlorované dibenzo-p-dioxiny/furany (PCDDs/Fs), polychlorované bifenyly (PCBs) či některé polycyklické aromatické uhlovodíky (PAHs). V testu jsem použila TCDD, který je nejsilnějším známým ligandem AhR. TCDD je karcinogenní látka, která může již v malých koncentracích způsobit širokou škálu toxických efektů. V teoretické části bakalářské práce jsem popsala toxické látky a jejich efekty, funkci Ah receptoru, mechanismy probíhající v buňkách, a princip stanovení aktivity luciferázy. V praktické části jsem popsala přesný postup práce s buňkami, použitý materiál a metody. V praktické části jsem nově zavedla linii RLT 2.0 a provedla řadu optimalizačních experimetnů. Získané výsledky jsem vyhodnotila (výpočet hodnoty EC50, násobku indukce a dalších parametrů ) a provedla srovnání s dalšími podobnými studiemi. Vypočítala jsem hodnoty EC50, které je možné využít pro další stanovení. Porovnala jsem také faktory, které mohou ovlivňovat indukci luciferázy. Na základě výsledků získaných při zavádění linie RLT 2.0 bude nyní možno používat linii RLT 2.0 pro rutinní stanovení AhR-zprostředkované aktivity dalších látek či vzorků ze životního prostředí.

Abstract:

In vitro biotests are suitable for examining the bioactivity of various components of the environment. In ecotoxicology bioassays with reporter genes are often used. The advantage of using these genes is their good sensitivity and reproducibility of results. There is also a simplicity factor to most of these tests. In this thesis I studied the activity of the Arylhydrocarbon receptor (AhR), which has a major role in the mediation of dioxin-like toxicity of chemicals. Binding to AhR causes biochemical changes in cells that lead to the regulation of gene transcription. In my experiments the RLT 2.0 recombinant cell line was used. RLT 2.0 is derived from rainbow trout hepatocarcinom which is stably transfected with pGudLuc1 plasmid. This plasmid contains a luciferase gene from the firefly. After exposure to the chemical compound, in this case 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), luciferase induction can be observed. In the presence of luciferase substrate a luminescent signal is generated. This signal is proportional to the AhR-mediated activity of tested compound. Dioxin-like toxicity express polychlorinated dibenzo-p-dioxins/furans PCDDs/Fs, polychlorinated biphenyls (PCBs) or certain polycyclic aromatic hydrocarbons (PAHs). In the bioassay I used TCDD, which is known as the strongest ligand of AhR. TCDD is a carcinogenic substance that can cause a wide range of toxic effects even in small concentrations. In the theoretical part of the thesis, the toxic substances, their effects, AhR function, the mechanisms proceed in cells, AhR activation and the principle of assesment of the luciferase activity are described. In the practical part, the RLT 2.0 cell line was newly introduced, and various optimizing experiments were performed. Obtained results were analyzed (EC50, fold-induction etc.), and the comparison with other similar studies was performed. Also the comparison of the factors that may affect the luciferase induction was examined. Based on the results obtained in the introduction of RLT 2.0 cell line, it will now be possible to use this cell line for routine determination of AHR-mediated activity.
