Bioassays for studies of specific toxicity in mixtures of environmental contaminants

Abstract:

Téma endokrinní disrupce je v popředí zájmu vědců již po několik desetiletí. Přírodní a umělé látky schopné narušovat endokrinní systém se v životním prostředí vyskytují ve složitých směsích, jejichž účinky lze jen obtížně předpovídat. In vitro biotesty umožňují zkoumat, zda jsou jednotlivé látky či komplexní směsi schopny vyvolat specifickou biologickou odpověď, včetně mechanismů endokrinní disrupce. Kombinace in vitro testů a analytických metod navíc umožňuje identifikovat látky vyvolávající tuto specifickou odpověď. Existují také in silico metody, které lze využít pro odhad škodlivých účinků chemických látek v organismu. Tato dizertační práce byla zaměřena na zkoumání specifických účinků směsí polutantů a přírodních látek ve vodním prostředí. K výzkumu bylo využito několik in vitro testů v kombinaci s analytickými metodami a modelováním. Práce obsahuje úvodní teoretickou část a přehled a shrnutí experimentálních výsledků ze tří samostatných studií. V přílohách jsou uvedeny vědecké publikace (I-IV), které obsahují podrobnosti diskutovaných studií. V první studii byly in vitro biotesty využity pro sledování dioxinové toxicity modelových směsí přírodní rozpuštěné organické hmoty s hydrofobními organickými kontaminanty (publikace I). Rozpuštěná organická hmota je hlavním zdrojem organického uhlíku ve vodě a je schopna interakce s mnoha kontaminanty. Ačkoli je rozpuštěná organická hmota schopna sorpce hydrofobních organických kontaminantů, což by teoreticky mělo vést ke snížení biodostupnosti a toxicity pro organismy, z výsledků naší studie vyplynulo, že organická hmota může v některých situacích zvýšit dioxinovou toxicitu kontaminantů. Druhá studie byla zaměřena na sledování dioxinových a endokrinně disruptivních účinků sedimentů (publikace II). Sedimenty akumulují mnoho hydrofobních organických kontaminantů, (xeno)hormonů a dalších endokrinních disruptorů a při remobilizačních procesech představují sekundární zdroj akumulovaných látek. Pro charakterizaci časové a prostorové variability v kontaminaci modelové oblasti dioxinovými, (anti)estrogenními a (anti)androgenními látkami byly využity celkem tři in vitro biotesty. Bylo zjištěno, že sezónní změny v říční hydrologie jsou důležitým faktorem ovlivňujícím kontaminaci sedimentů a je třeba je zohlednit při přípravě a interpretaci jakékoli monitorovací studie. Třetí studie byla zaměřena na zjištění hladin a účinků látek, které narušují glukokortikoidní a mineralokortikoidní signální dráhy (publikace III a IV). Tyto látky byly doposud studovány jen velmi okrajově, ovšem vzhledem k jejich rozsáhlému použití a zdrojům, zejména při léčebné terapii, mohou představovat významné endokrinní disruptory. Pro sledování glukokortikoidních účinků standardních látek a směsí látek extrahovaných ze vzorků odpadních a říčních vod byl použit GR-CALUX test. Výsledky z biotestů byly společně s výsledky chemické analýzy vzorků použity pro modifikaci již existující modelu pro přestup látek z vody do krevní plasmy ryb, tzv. „fish plasma model“, a pro odhad účinků směsí těchto látek v rybách. Závěrem lze shrnout, že výsledky získané v in vitro testech a v kombinaci s dalšími metodami významně přispěly k porozumění mechanismům interakce mezi přírodními a umělými látkami z mnoha různých skupin.

Abstract:

The field of endocrine disruption has been in the focus of researchers for the past few decades. Natural and synthetic compounds capable of disrupting the endocrine system are present in the environment in very complex mixtures and it is challenging to predict effects of these contaminated samples. In vitro bioassays enable to investigate potency of individual compounds as well as of complex mixtures to elicit specific biological responses, including mechanisms of endocrine disruption. Additionally, combination of both in vitro and instrumental methods enables the identification of compounds responsible for the specific response. Furthermore, various in silico methods can be used to extrapolate the potential of unintended effects of chemicals in organisms. The present thesis study aimed to investigate specific effects of mixtures of pollutants and natural compounds in the aquatic environment using various in vitro bioassays in combination with instrumental methods and modelling. The thesis consists of the introductory part and the overview of experimental results from three studies. Details on all the discussed studies can be found in annexes, where full texts of corresponding four research publications (I-IV) are provided. In the first study, dioxin-like toxicity of model mixtures of natural organic matter dissolved in the water (dissolved organic matter, DOM) with various hydrophobic organic contaminants (HOCs) was assessed using an in vitro assay (publication I). DOM represents a major source of organic carbon in water and many contaminants tend to associate with DOM. It was concluded that DOM does not seem to protect organisms against dioxin-like effects of HOCs as would be considered due to sorption of HOCs on DOM, but it can actually enhance their dioxin-like toxicity in some situations. In the second study, dioxin-like and endocrine disruptive potencies of sediments were investigated (publication II). Sediments accumulate many HOCs, (xeno) hormones and other endocrine disruptors and can provide a secondary source of contaminants during remobilization processes. Three different in vitro bioassays were successfully used to characterize temporal and spatial variability of contamination by dioxin-like, (anti)estrogenic and (anti)androgenic compounds in the model area. It was documented that the river hydrology with its seasonal changes is a very important parameter that needs to be taken into account both in design and interpretation of any monitoring study employing the effect-based assessment. Finally, the focus of the third part of the thesis was to generate novel information about levels and mixture effects of compounds that interfere with glucocorticoid and mineralocorticoid signalling (publications III and IV). These compounds have not been intensively studied, yet, however, they can be important environmental disruptors due to their extensive release, especially during medical treatment. The GR-CALUX bioassay was used to investigate glucocorticoid-like effects of mixtures extracted from wastewater and river water samples as well as to derive relative potencies of standard compounds to interact with the glucocorticoid receptor. Consequently, the bioassay derived data, together with the data from chemical analysis, were used to modify the existing fish plasma model and to extrapolate the mixture effects in fish. In conclusion, data obtained from in vitro bioassays, in combination with other methods, provided an important insight into the mechanisms of interactions among compounds of different classes.
