Profiling of membrane lipids associated with the pathogenesis of Alzheimer's disease

Abstract:

Alzheimer's disease is the most common type of dementia (60–80 %). It is widely spread especially in population over 65 years and its incidence is higher as age increasing. Diagnosis of Alzheimer's disease is very difficult and confidently determined only post-mortem. That is a reason why it is an immense effort to find new biomarkers and new reliable methods of her diagnosis preferably in the preclinical stage. The pathogenesis of Alzheimer's disease is closely related to the occurrence of gangliosides in the brain, which concentrations are changed during the life. Our aim was to optimise extraction method of gangliosides from serum and observe changes of concentration of gangliosides in the blood serum with regard to age, using ultra-high performance liquid chromatography and tandem mass spectrometry.

Abstract:

Alzheimerova choroba je najčastejším typom demencie (60-80 %). Je veľmi rozšírená najmä v populácii nad 65 rokov a so zvyšujúcim sa vekom stúpa jej incidencia. Diagnostika Alzheimerovej choroby je ná-ročná a s istotou sa dá stanoviť až pri pitve. Preto je veľká snaha o ná-jdenie nových biomarkerov a nové spoľahlivé metódy jej stanovenia, najlepšie už v prelinických štádiach ochorenia. S patogenézou Alzheimerovej choroby úzko súvisí zloženie a koncentrácia gagngliozidv v mozgu, ktoré sa počas života menia. Na-ším cieľom bolo optimalizovať metódu extrakcie gangliozidov zo séra a sledovať zmeny koncentrácii v krvnom sére ohľadom na vek, pomocou ultra-účinnej kvapalinovej chromatografie s tandemovou hmotnost-nou spektrometriou.
