Abstract:

Dizertační práce se zabývá patogenezí Alzheimerovy choroby. Alzheimerova choroba patří mezi závažná neurodegenerativní onemocnění a vede k úpadku tzv. kognitivních funkcí – myšlení, paměti, úsudku. Tato nemoc je nejčastější příčinou demence a v současné době není léčitelná. Existuje několik hypotéz o tom, jak Alzheimerova choroba vzniká, avšak celý proces patogeneze stále nebyl jednoznačně objasněn. V rámci této dizertační práce byly zanalyzovány jednonukleotidové polymorfismy několika kandidátních genů – TOMM40, CD36, CHAT a IDE, přičemž byly zjištěny statisticky významné asociace mezi určitými genetickými variantami studovaných genů a rizikem vzniku AD s pozdním nástupem. Snahou této práce je taktéž popsat, jak mohou proteinové produkty těchto genů ovlivnit vývoj Alzheimerovy choroby a jakým způsobem se pravděpodobně podílejí na interakcích s Aβ a tau proteiny, které patří k typickým znakům charakterizujícím patofyziologii AD.

Abstract:

The dissertation deals with the pathogenesis of Alzheimer’s disease. Alzheimer’s disease is a serious neurodegenerative illness and leads to the decline of cognitive functions – thinking, memory and judgment. This disease is the most common cause of dementia and currently is not treatable. There are several hypotheses of how Alzheimer’s disease arises, but the whole process of pathogenesis has still not been clarified. In this dissertation, single nucleotide polymorphisms of several candidate genes – TOMM40, CD36, CHAT, and IDE – were analyzed and it was found statistically significant associations between certain genetic variants of studied genes and the risk of developing the late-onset Alzheimer’s disease. The aim of this work is also to describe how could protein products of these genes influence the development of Alzheimer’s disease and how they are likely involved in interactions with Aβ and tau proteins, which are typical features of Alzheimer’s disease pathophysiology.
