The effect of selected Amaryllidaceae family alkaloids on viability and apoptosis induction in cancer cell lines

Abstract:

Tato diplomová práce se zabývá vlivem vybraných devíti alkaloidů čeledi Amaryllidaceae na viabilitu a indukci apoptózy u nádorových buněčných linií Jurkat a SK-BR-3. Alkaloidy rostlin představují perspektivní možnost terapie nádorových onemocnění, proto jsou v posledních letech předmětem testování v mnoha výzkumných laboratořích. Pro rozdělení skupiny alkaloidů, kterou jsme měli k dispozici, dle jejich cytotoxické účinnosti, byly použity dvě nezávislé metody. Nejprve byla použita spektrofotometrická end point analýza s tetrazoliovou solí XTT. Ve stejných koncentracích byly alkaloidy testovány metodou RTCA na přístroji xCELLigence, která zaznamenává změnu cell indexu buněk v čase. U všech alkaloidů byla vypočtena hodnota IC50 s ohledem na jejich potenciální využití pro účely protinádorové terapie. Dle výsledků z těchto experimentů byly tři nejúčinnější alkaloidy podrobeny dalším testům. Na průtokovém cytometru byla detekována změna buněčného cyklu ovlivněných buněk Jurkat a ve fluorescenčním mikroskopu byla pozorována indukce apoptózy vlivem alkaloidů. Testované alkaloidy se ve své cytotoxicitě velmi lišily, některé neměly na buňky žádný vliv a některé způsobovaly masivní umírání buněk apoptózou a inhibicí jejich proliferace.

Abstract:

This thesis describes the effect of selected nine alkaloids from Amaryllidaceae plant family on the viability and apoptosis induction in Jurkat and SK-BR-3 cancer cell lines. Plant alkaloids as a anticancer agens represents a perspective way of tumor therapy, therefore in the recent years are the subject of many anticancer research studies. For the classification of a examined group of alkaloids that were available for experiments based on their cytotoxic activity we used two independent methods. First we used spectrophotometric end point assay with tetrazolium salt XTT. Similarly, the same concentrations of alkaloids were tested using the RTCA on the xCELLigence to determine changes in cell index based on time following treatment. For all alkaloids IC50 value was calculated with aim to evaluate their potentional for use in anticancer therapy. According to the results obtained from these experiments three most effective alkaloids were selected for further testing. On the flow cytometer were determined the changes in the cell cycle distribution of the Jurkat treated cells and fluorescence microscopy was used to reveal apoptosis induction due to exposure to selected alkaloids. The studied alkaloids were very different in their anticancer activity. Some of them had no effect on cell viability and some caused massive cell death by apoptosis and inhibited proliferation of examined cells.
