Study of the biological activity selected isochinoline alkaloids

Abstract:

Izochinolinové alkaloidy jsou skupinou přírodních látek zahrnující několik skupin alkaloidů. Pro naše studium jsme vybrali kvarterní benzo[c]fenantridinové alkaloidy představující skupinu alkaloidů vykazujících široké spektrum zajímavých farmakologických účinků. Hlavními zdroji těchto alkaloidů jsou rostliny čeledí Papaveraceae, Fumariaceae, Ranunculaceae a Rutaceae, které jsou stovky let využívány v tradiční medicíně. Nejlépe prozkoumané kvarterní benzofenantridiny jsou sanguinarin a chelerytrin, které jsou komerčně dostupné. Vzhledem k tomu, že antiproliferační účinek sanguinarinu a chelerytrinu na nádorové buněčné linie byl popsán mnoha autory, jsou tyto alkaloidy diskutovány jako potenciální cytostatika. Předmětem studia této práce jsou v menší míře zastoupené minoritní benzofenantridiny sanguilutin, chelilutin, sanguirubin, chelirubin a makarpin, které jsou studovány málo. Jejich účinek na normální a nádorové buněčné linie byl porovnán se sanguinarinem a chelerytrinem. Antiproliferační aktivita alkaloidů byla hodnocena pomocí MTT testu. Všechny alkaloidy vykazovaly výraznou antiproliferační aktivitu na všech testovaných buněčných liniích bez ohledu na nádorový charakter linie. Hodnoty IC50 po 72 hodinové kultivaci se pohybovaly v rozmezí 0,012-1,58 µg/ml. Pro studium mechanismu indukce buněčné smrti byla vybrána nejcitlivější buněčná linie HL-60. Zaměřili jsme se na testování schopností kvarterních benzo[c]fenantridinových alkaloidů indukovat apoptózu a ovlivnit buněčný cyklus. Všechny alkaloidy byly výraznými induktory apoptózy, která byla potvrzena detekcí fragmentace jader pomocí DAPI, formací DNA žebříčků na agarózových gelech, elektronovou mikroskopií a průtokovou cytometrií buněk značených pomocí Annexinu V a propidium jodidu. Analýza buněčného cyklu prokázala přítomnost sub-G1 frakce po účinku sanguinarinu a chelerytrinu. Po účinku sanguilutinu, chelilutinu, sanguirubinu a chelirubinu byly patrné dva cykly naznačující aneuploidii. Značením pomocí propidium jodidu a metody TUNEL jsme prokázali, že nejde o aneuploidii, ale o apoptózu indukovanou u buněk ve všech fázích buněčného cyklu. Naše výsledky ukazují, že testované minoritní benzofenantridinové alkaloidy mají silný antiproliferační a proapoptotický efekt in vitro, které nejsou důsledkem oxidačního stresu vyvolaného alkaloidy. Naopak jsme stanovili antioxidační vlastnosti alkaloidů s výjimkou sanguilutinu 1 µg/ml a chelerytrinu 1 µg/ml. Práce představuje minoritní kvarterní benzofenantridinové alkaloidy jako látky zajímavé pro další výzkum v oblasti nádorové biologie.

Abstract:

Isochinoline alkaloids are natural substances comprising several different types of alkaloids. We studied quaternary benzo[c]phenantridine alkaloids which have a broad range of pharmacological effects. They are produced by plants from the families Papaveraceae, Fumariaceae, Ranunculaceae and Rutaceae and used for centuries in traditional medicine. The best known quaternary benzo[c]phenantridine alkaloids include sanguinarine and chelerythrine which have been studied in detail and which are also commercially available. Since sanguinarine and chelerythrine antiproliferative effects on cell lines have been reported by many authors, quaternary benzophenanthridines are discussed as potential cytostatic drugs. In this study we focused on five minor benzophenanthridine alkaloids namely sanguilutine, chelilutine, sanguirubine, chelirubine and macarpine, and compared their antiproliferative activities on normal and cancer cell lines with those of sanguinarine and chelerythrine. As assessed by the MTT test, all minor alkaloids showed significant antiproliferative activity on all cell lines tested, regardless of tumour or non-tumour origin. The IC50 values after 72 hours of cultivation ranged from 0.012 to 1.58 µg/ml. The mechanism of cell death induction was studied in the HL-60 as the most sensitive cell line. The ability of benzophenanthridine alkaloids to induce apoptosis and affect the cell cycle was tested. All alkaloids were strong inducers of apoptosis, which was demonstrated by the detection of nuclear fragmentation using DAPI staining, DNA ladders visualised by agarose gel eletrophoresis, electron microscopy and flow cytometry analysis of cells stained with Annexin V and propidium iodine. Cell cycle analysis showed the presence of sub-G1 fractions in cells exposed to the effect of either sanguinarine or chelerythrine. The cell lines treated with sanguilutine, chelilutine, sanguirubine or chelirubine had two cycles suggestive of aneuploidy. A double staining with propidium iodine and the TUNEL method excluded aneuploidy and confirmed apoptosis induced in all phases of the cell cycle. Our results show that the minor benzophenanthridine alkaloids tested have strong in vitro antiproliferative and proapoptotic effects that are not the results of alkaloid-induced oxidative stress. In contrast, we demonstrated antioxidative properties of some of these alkaloids except of sanguilutine 1 µg/ml and chelerythrine 1 µg/ml The study presents the minor quaternary benzo[c]phenantridine alkaloids as agents interesting in terms of further research into cancer biology.
