Cytogenetická a molekulárně-cytogenetická analýza chromosomových aberací u dětských pacientů s diagnózou non-Hodgkinův lymfom

Cytogenetic and molecular-cytogenetic analysis of chromosomal aberrations in childhood non-Hodgkins lymphomas

Abstract:

Malignant lymphomas are the third most common type of cancer in children. Non-Hodgkin´s lymphomas (NHL) represent heterogenous group of neoplasms, which develop from B or T lymphocytes and are the fastest growing childhood tumors. The field of cytogenetics significantly contributes to the diagnosis and identification of recurrent chromosomal rearrangements in hematological malignancies, which is a basic condition for identifying genes involved in the pathogenesis of lymphoma. In this study the cultivation of lymphoma cells was optimized and subsequently tumor and bone marrow samples from 21 pediatric patients with NHL were examined by cytogenetic and molecular-cytogenetic methods. Although there was a small set of samples analysed, the identified chromosomal abnormalities were used for further characterization of malignant lymphomas and for detection of influence of prognostically important cytogenetic aberrations on the clinical status of pediatric patients.

Abstract:

Malígne lymfómy sú po leukémiach a mozgových tumoroch tretím najčastejším nádorovým ochorením u detí. Non-Hodgkinské lymfómy (NHL) predstavujú rôznorodú skupinu neoplázií vychádzajúcich z lymfocytov radu B alebo T a patria k najrýchlejšie rastúcim nádorom vôbec. Odbor cytogenetika významne napomáha k diagnóze a identifikácii opakujúcich sa chromozómových prestavieb pri hematologických malignitách, čo je základnou podmienkou pre identifikáciu génov zahrnutých v patogenéze lymfómov. V tejto práci sa optimalizovala kultivácia lymfómových buniek a následne sa vyšetrili vzorky tumoru a kostnej drene 21 pediatrických pacientov s NHL pomocou cytogenetických a molekulárno-cytogenetických metód. Aj keď bol vyšetrovaný súbor vzoriek malý, identifikované chromozómové abnormality sa podarilo využiť k bližšej charakterizácii malígnych lymfómov a k zisteniu vplyvu prognosticky významných genetických zmien na klinický stav detských pacientov.
