Changes of membrane properties of epitelial cells in conditions of modulation of lipid metabolism

Abstract:

Epiteliální buňky mohou být transformovány v procesu karcinogeneze řadou chemických a virových agens. Tyto procesy jsou modulovány jednak cizorodými látkami (např. genotoxickými polyaromatickými uhlovodíky PAU), jednak endogenními látkami (cytokiny, hormony, lipidními signálními molekulami). V této práci byly epiteliální buňky exponovány vybranými cizorodými látkami (polychlorované aromatické látky), které ovlivňují fyzikální vlastnosti membrán, složení fosfolipidů a mastných kyselin a lipidní signální transdukci. Byly stanoveny změny hladin proteinů (β-katenin, β-aktivní-katenin, γ-katenin, E-kadherin, FAK, p-FAK, kaveolin-1 a kaveolin-2) pomocí Western blotting. Byla také stanovena fludita membrán po okamžitém účinku 25 µM a 40 µM PCB 153, 100 nM a 25 µM PCB 126, a po 1 a 6 hodinové expozici 40 µM PCB 153 a po 24 hodinové expozici 40 µM a 25 µM PCB 153 a DMSO jako negativní kontroly. V našem pokusu byl využit Luminiscenční Spektrometr LS 55 s xenonovou lampou a softwarem FL WinLab. Přístroj se dá využít k měření fluorescence, fosforescence a chemiluminiscence nebo bioluminiscence. Byla měřena intenzita fluorescence, polarizace fluorescence a anizotropie.

Abstract:

Epithelial cells can be transformed in a process of cancerogenity by various chemical and virus agents. These processes are modified by foreign materials (for example genotoxic PAU) and by endogenic materials (cytokinins, hormones, lipid signal molecules). In this article, epithelial cells were exposed to chosen foreign materials (polychlorinated aromatic materials), which influenced the physical characteristics of the membranes, the constitution of phospholipids and fatty acids and the lipid signal transduction. Following proteins were determined by Western blotting – β-catenin, active-β-catenin, γ-catenin, E-cadherin, FAK, p-FAK, caveolin-1 and caveolin-2. The fluidity of membranes was determined after a short-time effect of 25 μM and 40 μM PCB 153, 100 nM and 25 μM PCB 126 and after 1 and 6 hours exposure 40 μM PCB 153 and after 24 hours exposure 40 μM and 25 μM PCB 153 and DMSO as negativ control. In our experiment, we used Luminescent Spectrometer LS 55 with a xenon-lamp and FL WinLab software. The device can be used for measuring fluorescence, phosphorescence, chemiluminescent and bioluminescent. It was used for measuring the intensity of fluorescence, fluorescence polarization and anisotropy.
