Disagreement between morphological and molecular taxonomy of freshwater snails

Abstract:

Tradiční taxonomie plžů je založena zejména na morfologii jejich ulit. Druhy jsou také odlišovány pomocí znaků na měkkých tkáních, jako je anatomie pohlavní soustavy nebo pigmentace pláště. S využitím molekulárních znaků se však často ukazuje, že zejména morfologie ulit podléhá značné plasticitě v rámci jednoho druhu. Tato práce je rozdělena na rešeršní a praktickou část. V první části sumarizuji morfologické a molekulární znaky používané pro popisy druhů sladkovodních plžů. Dále vyhodnocuji nesoulad mezi vymezením druhů na základě morfologických znaků a rozdílů v sekvencích DNA. Praktická část se snaží najít způsob spolehlivého morfologického odlišení dvou mitochondriálních linií kružníka malého (Gyraulus parvus). Tyto dvě linie byly až do nedávna považovány za samostatné druhy: evropský kružník hlad-ký (Gyraulus laevis) a severoamerický G. parvus. Jelikož byla severoamerická linie zavlečena do Evropy, kde se rychle šíří a vykazuje větší ekologickou toleranci než evropská linie, je vhodné najít spolehlivé odlišovací znaky, jiné než molekulární. Možným kandidátem se jeví pigmentace tykadel. Z dat získaných molekulárními metodami a hodnocením pigmentace tykadel bylo cílem posoudit příslušnost jedinců kružníka malého k jedné ze dvou mitochondriál-ních linií. Výsledky v bakalářské práci existenci dvou linií potvrdily a přiřazení pigmentace tykadel většinově odpovídala tomuto rozdělení. Vybočující výjimky byly způsobeny výskytem přirozené variability v tomto znaku a pravděpodob-ným vznikem hybridních jedinců. Toto zjištění by v budoucnu mohlo mít prak-tické uplatnění v ochraně původních populací.

Abstract:

The traditional taxonomy of gastropods is based mainly on the morphology of their shells. Species are also distinguished by soft tissue characters such as genital anatomy or mantle pigmentation. However, using molecular charac-ters, it is often found that shell morphology in particular is subject to considerable plasticity within a species. This thesis is divided into a research and a practical part. In the first part, I summarize the morphological and molecular characters used to describe freshwater gastropod species. I also evaluate the discrepancy between species delineations based on morphological characters and differences in DNA sequences. The practical part attempts to find a way to reliably morphologically distinguish between the two mitochondrial races of Gyraulus parvus. These two races were until recently considered separate species: the European Gyraulus laevis and the North American G. parvus. Since the North American lineage has been introduced into Europe, where it is spreading rapidly and shows greater ecological tolerance than the European race, it is advisable to find reliable distinguishing features other than molecular ones. Tentacles pigmentation appears to be a possible candidate. From the data obtained by molecular methods and based on assessment of tentacle pigmentation I aimed to assess the affiliation of individuals of Gyraulus parvus to one of the two mitochondrial races. The results in the bachelor thesis confirmed the existence of two races and the assignment of tentacle pigmentation mostly corresponded to this distribution. The outliers were due to the occurrence of natural variation in this trait and probable existence of hybrid individuals. This finding could have an important application in the conservation of native populations.
