Role of cryptochromes in magnetoreception and circadian rythms of Periplaneta americana.

Abstract:

Předkládaná diplomová práce se věnuje uvažované úloze proteinu kryptochrom v mechanismech magnetorecepce a cirkadiánních rytmů živočichů se zvláštním zaměřením na švába amerického (Periplaneta americana). Práce je členěna na dvě hlavní části. První, teoretická část shrnuje dosavadní po-znatky oblasti endogenních cirkadiánních biologických hodin, jejich molekulární podstatu a zapojení klíčových molekul proteinů kryptochrom a timeless v různých živočišných druzích. Rovněž popisuje základní principy magnetické orientace i potenciální mechanismy magneto-recepčního smyslu a uvádí do problematiky tlumení genové exprese pomocí přirozeného obranného mechanismu RNA interference. Praktická část je zaměřena na behaviorální expe-rimenty cirkadiánní pohybové aktivity a magneticky indukovaného pohybového neklidu s použitím metod reverzní genetiky. Genetickými postupy byl získán originální výsledek svědčící pro funkci CRY jako receptoru citlivého na směrovou složku přirozeného magnetického pole. Naměřené výsledky přinášejí zatím jeden z nejpřesvědčivěj¬ších důkazů světlem indukované na kryptochromu za-ložené chemické magnetorecepce a historicky první evidenci zapojení CRY savčího typu v regulaci cirkadiánních rytmů hmyzu.

Abstract:

This master degree thesis deals with suggested role of cryptochrome protein in mag-netoreception and circadian rhythm mechanism of different animal species with special cov-erage on the American cockroach (Periplaneta americana). The thesis is divided into two main parts. First, theoretical part summarizes up the existing knowledge of the endogenous circadian biological clocks, its molecular principles and the role of key protein molecules of cryptochrome and timeless in different animal species. It further describes the essential principles of magnetic orientation, potential mechanism of magnetoreception and introduces the down regulation of gene expression issues, RNA in-terference. The practical part focuses at behavioral circadian activity experiments and at mag-netically induced movement restlessness using reverse genetics methods. Using genetic approaches we got an original result confirming the role of cryptoch-rome as a light-sensitive receptor with the response to the magnetic field direction. Our expe-rimental outcomes bring out one of the most convincing evidence of light induced cryptoch-rome based on the chemical magnetoreception and historical first evidence of the role of mammalian cryptochrome in insect circadian rhythm regulation.
