Interactions of beta-catenin and Ah receptor in regulation of beta-catenin-dependent transcription

Abstract:

Aktivace kanonické Wnt dráhy i indukce AhR hrají významnou roli v regulaci funkcí jaterních progenitorových buněk, které se podílejí na procesech jaterní regenerace a hepatokarcinogeneze. Cílem této diplomové práce bylo stanovit možné účinky TCDD na expresi genů, které jsou regulovány aktivací β-kateninu, samotného AhR a jeho cílových genů v buněčné linii WB-F344. Bylo potvrzeno, že aktivace kanonické Wnt dráhy pomocí myšího rekombinantního Wnt3a vede k aktivaci modelových cílových genů této dráhy – Axin2 a Ccnd1. Po působení Wnt3a byla také zvýšena hladina AhR a jeho modelových cílů – Cyp1a1 a Cyp1b1. Na základě výsledků studia globální genové exprese byla vybrána skupina genů, jejichž exprese může být regulována kanonickou Wnt dráhou. U všech těchto genů bylo zjištěno, že působení TCDD snižuje jejich expresi. Exprese genů Axin2, Ccnd1, Btrc, Cyp2e1, Efnb1, Fgf9 a Lef1 byla v použitém buněčném modelu zvýšena působením rekombinantního Wnt ligandu, což potvrdilo, že aktivace kanonické Wnt dráhy vede k indukci jejich transkripce. Naopak geny Bmp4, Edn1, Fn1 a Id2 nebyly v buňkách WB-F344 indukovány po aktivaci kanonické Wnt signalizace. Tento zdánlivý rozpor by mohl souviset s tím, že exprese některých genů regulovaných Wnt ligandy je tkáňově specifická a může být ovlivněna aktivitou dalších signálních drah. Byla také sledována exprese markerů jaterních progenitorových buněk, Ncam1, Scf, Krt14 a Krt19, která se v průběhu diferenciace jaterních progenitorových buněk do hepatocytů výrazně snižuje. Exprese těchto genů byla významně snížena působením TCDD, což naznačilo možné diferenciační působení AhR ligandů, jež bylo potvrzeno zvýšenou expresí Krt8, markeru hepatocytů. Negativní účinky TCDD na expresi cílových genů kanonické Wnt dráhy by mohly souviset s tím, že po působení TCDD dochází k degradaci aktivního β-kateninu. Chtěli jsme také ověřit, zda buňky WB-F344 mohou samy produkovat faktory aktivující β-katenin. Výsledky zatím nepotvrdily přítomnost faktorů aktivujících kanonickou Wnt dráhu v médiu z buněk WB-F344. Závěrem lze konstatovat, že TCDD v použitém buněčném modelu inhibuje bazální expresi řady genů, které jsou regulované kanonickou Wnt signální dráhou, a je také schopen potlačit expresi některých markerů jaterních progenitorových buněk. Tyto výsledky potvrzují, že aktivní β-katenin se hraje významnou reli v regulaci funkcí jaterních progenitorových buněk.

Abstract:

Activation of canonical Wnt signaling and AhR induction both play a significant role in regulation of functions of liver progenitor cells, which may participate in liver regeneration and hepatocarcinogenesis. The aim of this diploma thesis was to determine possible effects of TCDD on expression of the genes that are regulated by β-catenin activation, AhR and its target genes in WB-F344 cell line. It was confirmed that activation of canonical Wnt signaling pathway by mouse recombinant Wnt-3a leads to activation of model target genes of this pathway – Axin2 and Ccnd1. Also the levels of AhR and its model targets, Cyp1a1 and Cyp1b1, were increased after Wnt-3a treatment. Based on the results of global gene expression study we selected a group of genes, which expression can be controlled by canonical Wnt signaling pathway. It was found that TCDD treatment decreased expression of all of these genes. Expression of Axin2, Ccnd1, Btrc, Cyp2e1, Efnb1, Fgf9 and Lef1 genes was increased after recombinant Wnt ligand treatment in the cellular model used, which confirmed that the activation of canonical Wnt signaling pathway leads to induction of their transcription. On the contrary, Bmp4, Edn1, Fn1 and Id2 genes have not been induced after activation of canonical Wnt signaling in WB-F344 cells. This apparent discrepancy might be related to the fact that expression of some genes regulated by Wnt ligands is tissue specific and can be affected by activity of other signaling pathways. We also monitored expression of liver oval cell markers, Ncam1, Scf, Krt14 and Krt19, which decreases during the differentiation of hepatic progenitor cells into hepatocytes. Expression of these genes was significantly decreased after TCDD treatment, which indicated possible differentiating activity of AhR ligands, which was confirmed by increased Krt8 expression, a hepatocyte marker. The negative effects of TCDD on expression of target genes of canonical Wnt signaling might be related to observed degradation of active β-catenin after TCDD treatment. We also aimed to verify, whether WB-F344 cells may produce factors activating β-catenin pathway. However, the results did not confirm the presence of factors activating canonical Wnt signaling pathway in medium from WB-F344 cells. In conclusion, the present data suggest that TCDD inhibits, at least in the cellular model used, the basal expression of a number of genes which are regulated by canonical Wnt signaling pathway, and it is able suppress expression of several liver progenitor cells markers. These results confirm that active β-catenin plays an important role in regulation of function of liver progenitor cells.
