Účinky benzo(c)phenanthridinových alkaloidů na buňky maligního melanomu

The effects of benzo(c)phenanthridine alkaloids on malignant melanoma cells

Abstract:

Předkládaná disertační práce se zabývá mechanismem účinku pěti benzo[c]fenanthridinových alkaloidů (BA) - sanguinarinu (SA), chelerytrinu (CHE) a chelidoninu (CHLD), sanguilutinu (SL) a chelilutinu (CHL) - na buňky linií lidského maligního melanomu (MM). MM je nádorové onemocnění, jehož incidence má v lidské populaci trvale vzestupný trend. Léčba MM, zvláště pak jeho diseminované formy, je v důsledku rezistence na běžná cytostatika i radioterapii, velice obtížná. Identifikace nových přístupů a látek, které by měly terapeutický potenciál v léčbě MM, tedy zůstává důležitým cílem v oblasti výzkumu rakoviny. Zjistili jsme, že všechny studované alkaloidy vykazují výrazný antiproliferační efekt na buňky linií lidského MM. Přes výraznou podobnost molekulární struktury jednotlivé BA zprostředkovávají buněčnou smrt různými biochemickými drahami. Buněčná smrt indukovaná SA, CHE, CHLD a CHL je zprostředkována apoptózou, jejímž mechanismem je pravděpodobně snížení hladin antiapoptotických proteinů Bcl-xL, Mcl-1 a XIAP, zatímco morfologické a fyziologické znaky buněk ovlivněných SL odhalily, že na odpovědi buňky na přítomnost SL se podílejí procesy nekroptózy a autofagie. Nekroptóza jako RIP1-dependentní forma buněčné smrti nezávislá na aktivitě kaspáz, zatímco autofagie jako protektivní proces, jež má za následek oddálení smrti buňky. Přestože BA interagují s DNA, jejich účinek není závislý na aktivitě tumor supresorového proteinu p53. Výsledky předkládané práce ukazují na dvojí význam BA. (1) BA jsou látky s významným antiproliferačním potenciálem, které jsou schopny výrazně ovlivnit růst a životaschopnost buněk MM, a to včetně buněk vykazujících resistenci k běžným cytostatikům, jako jsou buňky s nefunkčním p53 a buňky nadprodukující antiapoptotický protein Bcl-2. (2) BA jsou látky vhodné ke studiu procesů buněčné smrti a autofagie a jejich vzájemného vztahu, což může vést k odhalení dalších terapeutických cílů v léčbě MM i jiných nádorů.

Abstract:

The present thesis deals with mechanism of action of five benzo[c]phenanthridine alkaloids (BAs), namely sanguinarine (SA), chelerythrine (CHE), chelidonine (CHLD), sanguilutine (SL) and chelilutine (CHL), on malignant melanoma (MM) cell lines. MM has become the cancer with an increasing incidence rate. Its inoperable metastatic form is difficult to treat because it is usually resistant to conventional chemotherapy, both radio- and chemotherapy. Therefore, search for new treatment attitudes and substances with antiproliferative activity towards melanoma are an actual issue in cancer research. Here we show that all studied BAs demonstrated considerable antiproliferative effects accompanied by induction of cell death. Despite the profound similarities in the molecular structures of individual BAs, it is clear that the mechanism associated with cell death induction is different for each alkaloid. SA, CHE, CHLD and CHL induce cell death mediated by apoptotic pathway, while SL induces RIP1-dependent non-apoptotic type of cell death, necroptosis, which is accompanied by autophagy induction. Apoptosis induced by BAs is probably mediated by reduction in the levels of antiapoptotic Bcl-xL, Mcl-1 and XIAP proteins. Concerning to the SL action, processes of necroptosis and autophagy are involved in the response of melanoma cells to this BA, but their functional roles are diverse. While necroptosis represents a form of programmed cell death, autophagy serves as prosurvival mechanism, which delays the cell death. Although BAs represent a group of agents that are able to interact with DNA, induction of cell death apparently does not seem to require p53 activity. This fact could lead to more efficient treatment for a large proportion of human cancer. Taken together, presented results show a double meaning of BA. (1) BA are compounds with a considerable antiproliferative potential significantly affecting the growth and viability of cells MM, including cells resistant to conventional cytostatic agents, such as cells with dysfunctional p53 and cells overexpressing antiapoptotic Bcl-2 protein, but also (2) substances suitable for the study processes of cell death and autophagy and interplay between them.
