Study of phytoestrogen level changes in ruminal fluid

Abstract:

Fytoestrogeny jsou polyfenolické sekundární metabolity rostlin. Teoretická část této práce je věnována zvláště podtřídě fytoestrogenů - isoflavonům, jejich struktuře, metabolismu, schopnosti vázat se na estrogenní receptory a účinkům na lidské zdraví. Praktická část se zabývá vývojem a validací metody pro stanovení isoflavonoidů a equolu v ruminální tekutině pomocí LC/MS-TOF a dále studiem zastoupení a koncentrace jednotlivých isoflavonů v bachorové tekutině přežvýkavců, do níž se mohou fytoestrogeny dostat trávicími procesy požitého krmiva s obsahem těchto látek. Jedním z výsledků diplomové práce je zavedení metody pro stanovení isoflavonoidů v ruminální tekutině, založené na extrakci analytů do ethylacetátu. Pomocí této metody byly stanoveny hladiny ve vzorcích reálné bachorové tekutiny: daidzin 0,0 ng/ml, glycitin 0,0 ng/ml, genistin 0,0-0,9 ng/ml daidzein 0,9-45,2 ng/ml, glycitein 0,02-1,3 ng/ml, genistein 0,0-1,6 ng/ml a equol 8,6-866,0 ng/ml.

Abstract:

Phytoestrogens are polyphenolic secondary metabolites of plants. The theoretical part of this thesis is devoted to subclass of phytoestrogens - isoflavones especially, their structure, metabolism, the ability to bind to estrogen receptors and effects on human health. The practical part deals with the development and validation of method for the determination of isoflavones and equol in ruminal fluid using LC/MS-TOF as well as study of representation and concentration of each isoflavone in ruminal fluid of ruminants, where phytoestrogens can get by digestive process of the ingested feed containing these substances. One of the results of the thesis is to introduce method for the determination of isoflavones in ruminal fluid based on the extraction of analytes into ethyl acetate. According to this method were determined levels in samples of the real ruminal fluid: daidzin 0,0 ng/ml, glycitin 0,0 ng/ml, genistin 0,0-0,9 ng/ml daidzein 0,9-45,2 ng/ml, glycitein 0,02-1,3 ng/ml, genistein 0,0-1,6 ng/ml a equol 8,6-866,0 ng/ml.
