Host-patogen interaction in relation to the genetic diversity in selected population of dogs

Abstract:

Pes představuje modelový druh pro studium lidských onemocnění. Přibližně 400 dědičných chorob je společných psům i lidem a některé z mutací nalezených u psů byly následně potvrzeny i u člověka. Díky absenci veterinární péče pod velkým tlakem patogenů a nulovému chovatelskému programu jsou vesničtí psi unikátní skupinou vhodnou ke studiu selekce řízené patogeny. V této práci jsem se zabývala genetickou diverzitou 150 keňských vesnických psů v mikrosatelitních lokusech, MHC a non-MHC kandidátních imunitních genech. Kandidátní geny kódující Toll-like receptory, NOS3, MD2, MyD88 a IL6 byly vybrány na základě jejich možné úlohy v obraně proti infekcím. Jako vhodné markery bylo nalezeno 16 jednonukleotidových polymorfizmů a byly vypracována metodyka pro jejich genotypizaci na populační úrovni. Jde zřejmě o první studii u psů využívající k populačním analýzám non-MHC imunitní geny. Vstupní populace byla rozdělena na tři sub-populace. Získaná data naznačují, že toto genetické rozdělení vzniklo jako důsledek geografických a sociokulturních faktorů typických pro danou oblast. Některé z jednonukleotidových markerů byly asociovány s patogeny, jmenovitě se psím psinkovým virem, Neospora caninum, Hepatozoon canis a filáriemi. Výsledky naznačují, že především MyD88 – závislá signální dráha může hrát důležitou roli v mechanizmech rezistence k několika infekcím v této populaci.

Abstract:

Dogs represent a model species for studying human disease. They share approximately 400 hereditary diseases with humans and some mutations found in dogs were subsequently confirmed in humans. Due to the absence of veterinary care under strong pathogen pressure and with no selective breeding, country dogs are a unique group well suited for studies of pathogen - driven selection. In this thesis, I studied genetic diversity of 150 Kenyan dogs in microsatellite, MHC and selected non-MHC candidate immunity – related loci. The candidate genes encoding Toll-like receptors, NOS3, MD2, MyD88, and IL6 were chosen on the basis of their possible role in the defence against infections. Altogether, 16 single nucleotide polymorphisms have identified as suitable molecular markers and methods of their genotyping at the population level were developer. This is probably the first study reporting on genetic diversity of non-MHC immune genes in dogs. The genetic diversity of Kenyan village dogs was similar to the diversity of other village dogs studied. The entire population was shown to be stratified into three sub-populations. The data obtained suggested that the genetic stratification is due to a combination of geographical and sociocultural factors specifi for the region. Several immunity-related SNP markers were associated with infection, namely with canine distemper virus, Neospora caninum, Hepatozoon canis and filarias. The results suggested that aspecially the MyD88 pathway could play an important role in the mechanisms of resistence to the multiple infections in this particular population.
