Regulatory network of desmocollin-1 in the relationship with breast cancer metastasis

Abstract:

Desmocollin-1 (DSC1) je protein zapojený do stavby desmozomálních spojů, čímž zprostředkovává buněčnou adhezi. U DSC1 byla v předchozích studiích zjištěna spojitost s promigrační úlohou u nádorů prsu luminální A. Vzhledem k tomu byl také studován vliv inhibitorů na hladiny DSC1, přičemž parthenolid jako jediný signifikantně snižoval hladinu DSC1. Cílem práce bylo studovat vliv zvýšené exprese DSC1 na transkriptomický profil stabilně transdukované buněčné linie MCF7-DSC1-GFP za využití RNA sekvenování nové generace a dále sledovat i vliv parthenolidu. Získaná transkriptomická data byla hodnocena společně s daty na úrovni proteomu, přičemž v odpovědi na overexpresi DSC1 bylo identifikováno 31 významně deregulovaných genových produktů, mezi nimiž byly proteiny zapojené do proliferace buněk (USP8, IGFBP5, TOM1L1, FAM83B). Dále byla provedena GSEA pathway analýza spojených transkriptomických a proteomických dat. K obohacení drah došlo zejména u APICAL JUNCTION spojenou s rolí DSC1 v desmozomech a E2F TARGETS, která obsahuje geny spojené s regulací buněčného cyklu, transkripcí a proliferaci. Práce na transkriptomové úrovni potvrzuje souvislost overexprese DSC1 se zvýšením buněčné proliferace.

Abstract:

Desmocollin-1 (DSC1) is a protein involved in the construction of the desmosomal junctions, thereby mediating cell adhesion. In previous studies, DSC1 was found to be associated with a pro-migratory role in luminal A breast tumors. Therefore, the effect of inhibitors on DSC1 levels was also studied, with parthenolide being the only one significantly reducing DSC1 levels. The aim of the thesis was to study the effect of DSC1 overexpression on the transcriptomic profile of the stably transduced cell line MCF7-DSC1-GFP using next-generation RNA-sequencing and to further analyze the effect of parthenolide. The obtained transcriptomic data were evaluated together with data at the proteome level. In response to DSC1 overexpression, 31 significantly deregulated gene products were identified, including proteins involved in cell proliferation (USP8, IGFBP5, TOM1L1, FAM83B). Furthermore, GSEA pathway analysis of the combined transcriptomic and proteomic data was performed. Pathway enrichment occurred mainly in APICAL JUNCTION associated with the role of DSC1 in desmosomes and E2F TARGETS, which contain genes associated with cell cycle regulation, transcription and proliferation. The thesis confirms at transcriptome level the association of DSC1 overexpression with the increased cell proliferation.
