Possibilities of affection of the adherence and growth of cariogenic streptococci

Abstract:

Zubní kaz je infekční onemocnění s vysokou incidencí. Zásadní roli v etiologii zubního kazu hraje bakterie Streptococcus mutans. Ovlivnění kariogenního potenciálu a prevalence této bakterie v dutině ústní jsou předmětem zájmu řady in vitro a in vivo studií. Jednou z možností, která je v současnosti zvažována, je podávání probiotik – nepatogenních a netoxických druhů mikroorganismů, které při podávání v dostatečném množství přináší zdravotní prospěch svému hostiteli a jejich pozitivní účinek byl prokázán především v gastrointestinálním traktu. Tato práce se zabývala vlivem probiotických laktobacilů na růst a adhezi Streptococcus mutans in vitro. V první části testování, zaměřené na ovlivnění růstu S. mutans, byly provedeny dva testy. Test inhibice růstu sloužil k detekci inhibiční schopnosti jednotlivých kmenů laktobacilů, která byla potvrzena téměř u 70 % kmenů. Všechny kmeny s nejlepší schopností inhibice patřily do druhu Lactobacillus plantarum. Doplňujícím testem byl Agar Overlay Interference Test, kterým byla inhibiční schopnost vybraných kmenů laktobacilů opět potvrzena. V druhé části testování byla zjišťována inhibice tvorby biofilmu S. mutans pomocí buněčných produktů laktobacilů. V rámci testování byly použity tři různé směsi – buněčný lyzát, supernatant a směs lyzátu se supernatantem a zároveň byly vyzkoušeny tři různé postupy – pre-coating a souběžná kultivace ve dvou ředěních (20:80 a 40:60). Výraznou inhibiční schopnost měly všechny tři směsi, a to při použití všech postupů. Rozdíly byly patrné mezi jednotlivými postupy – až na jednu výjimku se prokázal výrazně nižší inhibiční vliv při použití postupu pre-coating. Co se týká jednotlivých kmenů, byl v porovnání s ostatními kmeny zaznamenán méně výrazný inhibiční efekt u kmene Lactobacillus helveticus CCM 7193T. Odlišnosti ve schopnosti inhibice byly patrné zejména u kmene Lactobacillus rhamnosus CCDM 579/02, který se v porovnání s dalšími kmeny v jednom případě ukázal jako nejvýraznější inhibitor, zatímco ve dvou dalších případech byly jeho inhibiční vlastnosti nejméně výrazné.

Abstract:

Dental caries is an infectious disease with a high level of incidence. Bacterium Streptococcus mutans is the most important aetiological agent for the development of dental caries. A lot of in vitro and in vivo studies attempt to influence the cariogenic potential and prevalence of this bacterium in oral cavity. Recently, the use of probiotics is point of great interest. Probiotics are nonpathogenic and nontoxic microorganisms which when administered in adequate amounts confer health benefits for the host. This thesis dealt with the influence of probiotic lactobacilli on the growth and adherence of S. mutans in vitro. The first part of the study was focused on the affection of growth and two tests were performed in this part. The growth inhibition test was used for detection of inhibition capacity of Lactobacillus strains, which was confirmed in almost 70 % of strains. All strains with the best inhibition capacity belonged to species Lactobacillus plantarum. The second Agar Overlay Interference Test was additional test by which the inhibition capacity of certain strains was approved again. In the second part of the study the inhibition of biofilm formation of S. mutans by cell products of lactobacilli was investigated. There were used three different mixtures (lysate, supernatant and lysate with supernatant) and three different types of cultivation (pre-coating and co-incubation in two proportions – 20:80 and 40:60). All three mixtures, as well as, all three types of cultivation showed high inhibition capacity. Noticeable differences were among particular types of cultivation. Significantly lower inhibition effect was proved in all assays using the pre-coating cultivation except for one. With respect to different strains, the Lactobacillus hellveticus CCM 7193 showed a weaker inhibition when compared to the others. Contradictory results were observed in Lactobacillus rhamnosus CCDM 579/02 – in one case it was the best inhibitor, meanwhile in two other cases it showed weaker inhibition capacity.
