Involvement of hydrogen sulfide in tumor fate

Abstract:

Dizertační práce se zabývá vlivem sirovodíku (H2S), jako třetího gasotransmiteru, na kolorektální, ovariální a světlobuněčný renálního karcinom (ccRCC). Experimenty byly prováděny na stabilních buněčných liniích in vitro a též in vivo na modelu imunodeficitní nahé athymické myši. Modulace hladiny sirovodíku byla navozena použitím exogenního donoru H2S - GYY4137 a též ovlivněním exprese/aktivity intracelulárních enzymů produkujících endogenní H2S. Studovány a diskutovány jsou účinky H2S na apoptózu zejména ve vztahu k nitrobuněčnému pH a intracelulárnímu kalciu, ale i možná role nitrobuněčných enzymů produkujících H2S v rezistenci světlobuněčného renálního karcinomu vůči chemoterapii.

Abstract:

The dissertation thesis deals with the effect of hydrogen sulfide (H2S), as the third gasotransmitter, on colorectal, ovarian and clear cell renal carcinoma. The experiments were performed in vitro on stable cell lines as well as in vivo using the immunodeficient nude athymic mouse model. For the modulation of the hydrogen sulfide level, the exogenous H2S donor - GYY4137 and also the modulation of the expression / activity of H2S-producing intracellular enzymes were utilized. The effects of H2S are studied and discussed in relation to apoptosis, particularly in connection with intracellular pH and intracellular calcium, but also a possible role of H2S-generating intracellular enzymes in the resistance of clear cell renal carcinoma to chemotherapy.
