V této práci jsme zkoumali funkce a mechanismy, pomocí kterých je TRAMP zapojen do transkripce, zpracování a kontroly kvality RNA v Saccharomyces cerevisiae. Úspěšně jsme odhalili, jak se jednotlivé podjednotky skládají do komplexu, jak TRAMP rozezná a váže molekuly RNA a nastínili jsme roli domény KOW v proteinu Mtr4p při aktivaci exozomu. Abstract:
In this thesis, we have investigated the function and mechanisms by which is TRAMP involved in RNA transcription, processing and quality control in Saccharomyces cerevisiae. Briefly, we successfully uncovered a puzzle of the RNA binding and assembly of the TRAMP complex and suggest a role of the KOW domain of Mtr4p in the exosome activation.
