Trichloroethene degradation at low temperature and other less favourable conditions

Abstract:

Principem kometabolické degradace je rozložit cizorodou látku za specifických podmínek, kdy pomocí enzymů, potřebných pro rozklad růstového substrátu, dochází k přeměně nerůstového substrátu. Cílem této diplomové práce bylo zjistit, zda jsou dříve získané bakteriální kultury, rostoucí na fenolu při 8 °C a současně při pH 5,2 nebo při zvýšené koncentraci NaCl, schopné degradovat trichlorethylén (TCE). Ověřovací růstové testy bakteriálních kultur 1AZS a 5BZS byly úspěšné téměř ve všech podmínkách prostředí, tedy při neutrálním i mírně kyselém pH, teplotách 8 °C i 23 °C a také v přítomnosti laktátu i bez něj. Jako poměrně úspěšná se ukázala degradace TCE v koncentraci 1,8 mg/l kulturou 5BZS, rostoucí na fenolu v kombinaci s laktátem, kdy bylo rozloženo při teplotě 8 °C a pH 5,2 téměř 53 % TCE, při teplotě 23 °C pak 62 %. Degradace za stejných podmínek prostředí a při snížené vstupní koncentraci TCE (1,2 mg/l), pH 5,2 a teplotě 8 °C byla úspěšná téměř z 90 %. Degradační testy v přítomnosti 4 % NaCl však příliš úspěšné nebyly. Kultury 10AZS a 8AZS byly i přes svůj dobrý růst schopny rozložit pouze necelých 30 % a 20 % TCE.

Abstract:

The principle of cometabolic degradation is to break down xenobiotic substances under specific condition, where enzymes responsible for the degradation of the growth substrate concurrently transform non-growth substrates. The aim of this thesis was to determine whether previously obtained bacterial strains, growing on phenol at 8°C and simultaneously at pH 5.2 or with increased NaCl concentration, are capable of degrading trichloroethylene (TCE). Verification growth tests of bacterial strains 1AZS and 5BZS were successful under almost all environmental conditions, including neutral and slightly acidic pH, temperatures of 8°C and 23°C, and in the presence or absence of lactate. Degradation of TCE at a concentration of 1.8 mg/l by strain 5BZS, growing on phenol in combination with lactate, proved relatively successful, with nearly 62% of TCE degraded at a temperature of 8°C and pH 5.2, as well as at 23°C. Degradation under the same environmental conditions and at a reduced input concentration of TCE (1.2 mg/l), pH 5.2, and temperature of 8°C was successful and nearly 90% of TCE was removed. However, degradation tests under increased NaCl concentration were not very successful. Strains 10AZS and 8AZS, despite their good growth on phenol in the presence of 4% NaCl and TCE, were able to degrade less than 30 % and 20 % of TCE.
