Chicken and hen immune response to the infection with Salmonella enterica

Abstract:

Rod Salmonella enterica patří mezi nejinvazivnější střevní patogeny, kteří mohou infikovat člověka a široké spektrum zvířat. Výsledný průběh a podoba infekce je určena interakcí konkrétního hostitele a serotypu. Infekce Salmonella enterica je u drůbeže doprovázena, v rámci prvotní odpovědi imunitního systému, expresí prozánětlivých cytokinů a chemokinů. Ve své práci jsem se zabývala indukcí cytokinů ve slepém střevě 39 denních kuřat plemene Lohmann Brown 4 dny po orální a intravenózní infekci kmenem Salmonella enterica subsp. enterica sv. Enteritidis 147. Kvantifikací mRNA pomocí Real-time PCR jsem určila příslušné hladiny cytokinů (IL-1β, IL-6, IL-8, IL-17, IL-18, IL-22, TNF-α, IFN-γ, iNOS). Nejvyšší exprese byla u neinfikované kontrolní skupiny u TNF-α jehož exprese byla více než dvojnásobná v porovnání s expresí referenčních house keeping genů. Po infekci salmonelami došlo u všech sledovaných cytokinů ke zvýšení exprese, přičemž ve všech případech byla indukce cytokiny vyšší po intravenózní infekci než po per orální. Po orální infekci došlo k nejvyšší indukci u IL-6, jehož exprese se zvýšila téměř 26x. Po intravenózní infekci došlo k nejvyšší indukci u IL-6, IL-8, IL-17,IL-22, IFN-γ a iNOS, jejichž exprese se zvýšila téměř 30 až 80x. Zjištěné údaje jsem porovnávala s výsledky, ke kterým dospěli dříve jiní autoři a zamýšlela jsem se nad důvody odlišností a shod mezi nimi.

Abstract:

The genus Salmonella enterica belongs to the most invasive intestinal patogens, that can infect human and wide range of animals. The consequentional process and form of infection are determined by the interaction of specific host and serotype. The infection of Salmonella enterica is accompanied, within the frame of primary response of the immune system, by the expression of pro-inflammatory cytokines and chemokines. In my work I was focused on induction of cytokines in the caecum of 39 days old chikens, belonging to Lohmann Brown breed, 4 days after oral and intravenous infection of genus Salmonella enterica subsp. enterica sv. Enteritidis 147. I appointed the levels of individual cytokins (IL-1β, IL-6, IL-8, IL-17, IL-18, IL-22, TNF-α, IFN-γ, iNOS) by mRNA quantification using Real-time PCR. The highest expression was present in the uninfected control group of Chickens TNF-α , and was more than double compared to reference housekeeping gene expression. After the infection of Salmonella the expression in all measured cytokines increased, whereas the induction of cytokines was higher after the intravenous infection than after the per oral in all of cases. The highest induction of IL-6 was after the oral infection. Its expression increased almost 26x. The highest induction of IL-6, IL-8, IL-17, IL-22, IFN-γ and iNOS was after the intravenous infection, the expression increased almost nearly 30-80x. I compared the observed data with the earlier results and I tried to explain the reasons for differences and similarities between them.
