Identification and characterization of proteins regulated by the plant hormone cytokinin

Abstract:

Rostlinné hormony cytokininy (CK) regulují velkou škálu významných procesů růstu a vývoje rostlin. I když jsou intenzivně studovány již řadu let, molekulární podstata jejich účinků stále není zcela objasněna. Proteomická analýza, která je hlavní náplní této práce, byla doposud v této oblasti značně opomíjena. Sledování účinku exogenní aplikace cytokininů (zastupujících cytokininy hydroxyisopentenylového typu, isopentenylového typu, aromatického typu a cytokininy močovinového typu) na úrovni proteomu a fosfoproteomu odhalilo celkem 96 proteinů zapojených do brzké odpovědi na cytokininy. Bližší zkoumání jejich možné role ukázalo mimo jiné na zapojení signalizace pomocí iontů vápníku a propojení mezi vnímáním teploty a signální dráhou cytokininů. Obě tato spojení byla prokázána následnými experimenty. Studium zapojení cytokininů do teplotní signalizace navíc odhalilo 148 proteinů, které jsou ovlivněny v odpovědi na 15 min teplotního šoku. Ukázalo se, že více než 70 % těchto proteinů je ovlivněno přítomností exogenních cytokininů, které do jisté míry napodobují účinky šoku zvýšenou teplotou. Důležitost cytokininů pro vnímání zvýšené teploty byla také prokázána fyziologickým experimentem.

Abstract:

Cytokinins (CKs) regulate diverse developmental processes in plants. To get an insight into cytokinin regulated molecular events at the proteome level, early cytokinin responses were investigated through proteome-wide expression profiling based on image and mass spectrometric analysis of two-dimensionally separated proteins and phosphoproteins. The effects of 15 min treatments of 7-day-old Arabidopsis thaliana seedlings with four main cytokinins representing hydroxyisopentenyl, isopentenyl, aromatic, and urea-type cytokinins were compared to help elucidate their common and specific function(s) in regulating plant development. In these experiments, 96 proteins were identified by MALDI-TOF/TOF mass spectrometry, providing a snapshot of early links in cytokinin regulated signaling. Novel targets of cytokinin signaling pointed to a possible link between temperature and cytokinin signaling and also the involvement of calcium ions in cytokinin signaling cascades. The connections were proven correct in consecutive experiments, which included the first proteomic analysis of 15 min temperature-shock responses in plants on the phosphoproteoeme, and proteome fractions (i) enriched in low abundance proteins via proteome equalization and (ii) depleted of RuBisCO. In this second large-scale experiment, 148 shock-response proteins were identified. More than 70 % of these temperature-shock response proteins were affected by exogenous cytokinin treatment which seems to partly mimic the effects of heat-shock on the Arabidopsis proteome. Further, a role for cytokinin in heat perception was proved in physiological experiments.
