The influence of diet on the composition of the gut microbiome

Abstract:

Tato bakalářská práce se zabývá střevním mikrobiomem a vlivem stravy na jeho složení. Práce obsahuje obecné informace o metodách výzkumu střevního mikrobiomu a jeho souvislostech se zdravím člověka. Zabývá se také tématem fekální mikrobiální transplantace, která sama o sobě demonstruje význam samotného střevního mikrobiomu a jeho vlivu na fenotyp člověka a zmiňuje možnosti jeho terapeutického využití. Hlavní část této práce je soustředěna na stravu jako celek, ale také na její jednotlivé složky a na jejich vliv na mikrobiální složení a enterotypy (rozdílné typy střevních mikrobiomů vzniklých na základě rozdílného dlouhodobého stravování). Bylo zjištěno, že enterotyp s převahou rodu Prevotella souvisí spíše s příjmem rostlinné stravy, zatímco typ Bacteroides je spojen spíše s příjmem živočišných proteinů a tuků. Enterotyp Prevotella je, zejména v kontextu západní civilizace, spojován s mnoha prospěšnými vlastnostmi, jako je například zlepšení profilu kardiovaskulárních rizikových faktorů a metabolismu glukózy. Ve studiích se také opakoval poměr kmene Firmicutes ku Bacteroidetes, který je při západním stravování zvýšený a bývá spojen s negativními účinky na lidské zdraví. Dále je také rozebrán vliv krátkodobých a dlouhodobých změn ve stravování na střevní mikrobiom s tím, že pouze dlouhodobé stravovací změny mají vliv na změnu složení. Práce je zakončena globálním přehledem složení střevního mikrobiomu a tím, jak mikrobiální složení odráží rozdílný typ stravování a životního stylu. Z práce vyplývá, že nejzdravější stravou je tzv. středomořský typ stravování.

Abstract:

This bachelor thesis deals with the gut microbiome and the influence of diet on its composition. The thesis contains general information about methods of research of the gut microbiome and its connections with human health. It also deals with the topic of fecal microbial transplantation, which itself demonstrates the importance of the gut microbiome itself and its influence on the human phenotype and mentions the possibilities of its therapeutic use. The main part of this thesis is focused on the diet as a whole, but also on its individual components and their influence on the microbial composition and enterotypes (different types of gut microbiomes created on the basis of different long-term diets). It was found that the enterotype dominated by the genus Prevotella is rather associated with the intake of plant-based foods, while the Bacteroides type is linked more to the intake of animal proteins and fats. The Prevotella enterotype is, especially in the context of Western civilization, associated with many beneficial properties, such as improving the profile of cardiovascular risk factors and glucose metabolism. In studies has also repeated the ratio of Firmicutes to Bacteroidetes, which is increased in Western diets and is often associated with negative effects on human health. Furthermore, it is also analyzed the influence of short-term and long-term changes in diet on the gut microbiome, with the understanding that only long-term dietary changes have an effect on the change in composition. The thesis is concluded with a global overview of the composition of the gut microbiome and how microbial composition reflects different types of diet and lifestyle. The thesis shows that the healthiest diet is the so-called Mediterranean type of diet.
