Monitoring of malondialdehyde concentration during plant defence reaction

Abstract:

Jako každý organismus i rostliny jsou během svého života vystavovány vlivům, které je mohou ohrozit a dokonce vést až k jejich odumření. Vyvinuly si proto obranné mechanismy, kterými brání průniku a rozšíření patogenního organismu v rostlině. Důležitou roli ve vztahu patogenu a rostliny zastávají elicitory. Elicitory jsou schopny vyvolat produkci aktivních forem kyslíku a zvýšenou expresi lipooxygenas, čímž je započata peroxidace lipidů. Tento velmi destruktivní proces zapříčiňuje poškození biologických membrán, což vede ke změně jejich funkce. Během peroxidace lipidů vzniká velké množství produktů, které dále mohou narušovat i jiné molekuly v organismu. Mezi nejvíce zastoupené metabolity patří malondialdehyd, který bývá označován jako marker oxidačního poškození organismu. Hladina malondialdehydu se nejčastěji určuje reakcí s thiobarbiturovou kyselinou a následnou analýzou pomocí HPLC. Cílem této diplomové práce je stanovení hladiny malondialdehydu v listech tabáku, jehož zvýšená produkce je vyvolána kryptogeinem a jeho mutantními proteiny (56, 56+R, L19R, L19R(EAEA), L41F, V84F, L41F/V84F, L80F/V84F), a současně stanovení prahové koncentrace jednotlivých proteinů schopné vyvolat zvýšenou produkci malondialdehydu. Výsledky prokázaly nekrotickou aktivitu proteinů 56, L19R, V84F a L80F/V84F. Jejich prahová koncentrace pak byla stanovena na hodnotu 2,5 nM. Naopak proteiny 56+R, L19R(EAEA), L41F a L41F/V84F neprojevovaly nekrotickou aktivitu ani při nejvyšších použitých koncentracích.

Abstract:

During their life, plants – just as any other organisms – are subject to influences that may endanger them and even lead to their necrosis. Therefore, they developed defense mechanisms with which they bar the pathogen organism from entering and spreading within the plant. An important role in the relationship between a pathogen and a plant is played by elicitors. Elicitors are capable of eliciting production of active oxygen species and increased expression of lipoxygenases by which the lipid peroxidation is started. This highly destructive process causes damage in biological membranes, leading to a change in their function. During lipid peroxidation, large amount of products that may further disturb other molecules in the organism is created. Among the most represented metabolites belongs malondialdehyde, which is often described as the marker of the organism’s oxidation damage. The level of malondialdehyde is usually determined through reaction with thiobarbituric acid and subsequent analysis with HPLC. The aim of this diploma thesis is to determine the level of malondialdehyde in tobacco leaves, its increased acumulation being caused by cryptogein and its mutant proteins (56, 56+R, L19R, L19R(EAEA), L41F, V84F, L41F/V84F, L80F/V84F). The threshold concentration of individual proteins able to induce the production of malondialdehyde is also established. Results proved necrotic activity of the 56, L19R, V84F and L80F/V84F proteins. Their threshold concentration was then determined at 2,5 nM. By contrast, the 56+R, L19R(EAEA), L41F and L41F/V84F proteins did not show necrotic activity, not even when the highest concentrations were used.
