Regulation of FGF signaling in chondrocytes

Abstract:

Signální dráha FGF má v buňce vliv na mnoho důležitých funkcí, jako je buněčná proliferace, diferenciace, přežití či migrace. Je klíčovou drahou nejen v embryonálním vývoji, ale i v dospělém organizmu. Cílem této práce je zkoumání vlivu chemických inhibitorů a genetického modifikování FGF signalizace v buňkách chrupavky. U chemických inhibitorů, statinů, které se běžně využívají ke snížení hladiny cholesterolu, bylo nedávno prokázáno, že působí proti aberantní FGFR3 signalizaci u buněk a zvířecích modelů s chondrodysplazií. Tohoto vlivu by se proto dalo využít například při léčbě trpaslictví u lidí. Mechanizmus účinku statinů na FGFR3 je stále nejasný, proto je v této diplomové práci zkoumán vliv čtyř statinů na signalizaci FGF v buňkách chrupavky. Pro práci byly využity techniky, které byly již dříve popsané jako vhodné na prokázání funkce inhibitorů, i přesto však statiny žádný přímý vliv na FGF signalizaci neprojevily. Dále bylo zkoumáno ovlivnění FGF signální dráhy modifikací genomu za využití CRISPR-Cas9 systému, díky kterému byla indukována ztráta funkce genu pro adaptérový protein FRS2 v buňkách chrupavky. Buňky bez tohoto proteinu však nebylo možné získat. Vzhledem k dosavadním zkušenostem s úspěšným targetingem jiných genů bylo usouzeno, že je protein FRS2, díky své účasti i v dalších signálních drahách, pro chondrocyty nezbytný a buňky bez něj nedokáží přežít.

Abstract:

FGF signaling pathway influences a number of important functions such as cell proliferation, differentiation, survival and migration. It is a key pathway in the embryonal development as well as in the mature organism. The aim of the thesis is to examine the effect of chemical inhibitors and genetic modifications of the FGF signaling in chondrocytes. In case of the chemical inhibitors, statins, which are commonly used to lower the level of cholesterol it has been proved recently that they counteract aberrant FGFR3 signaling in cells and animal models with chondrodysplasia. This effect could be used for example in the treatment of dwarfism in humans. The mechanism of the effects of statin on the FGFR3 signaling in cells has not still been clarified, therefore the effect of four statins on the FGF signaling in cartilage cells is studied. Techniques that were previously described as appropriate to show the functions of inhibitors were used, however statins did not show any direct influence on the FGF signaling. Further, the effect on the FGF signaling pathway using the genome modification with CRISPR-Cas9 system was applied. Due to which the loss of gene function for the FRS2 adapter protein in chondrocytes was induced. Cells without this protein were not able to obtain. Due to the up-to-date experience with positive targeting of other cells, it has been estimated that the FRS2 protein, thanks to its presence also on other signal pathways, is essential for chondrocytes and cells cannot live without it.
