Triclosan and triclocarban: environmental occurrence, toxicity and modes of action

Abstract:

Triclosan a triclocarban jsou syntetické antimikrobiální látky hojně užívané v produktech osobní péče, čisticích přípravcích, humánních a veterinárních léčivech a v průmyslu. V důsledku jejich nadměrného užívání a nedokonalého odstraňování při čištění odpadních vod vstupují tyto látky do prostředí, kde jsou detekovány jak v povrchových vodách, sedimentech či půdách, tak ve vodních i suchozemských organismech včetně lidských tkání. Přestože nejsou akutně toxické, mutagenní ani teratogenní pro savce, můžou mít tyto látky nepříznivý vliv na ostatní organismy, obzvlášť při jejich dlouhodobém působení. Mezi jejich mechanismy účinku patří narušování hormonální regulace a signálování, což je řadí mezi tzv. endokrinní disruptory. Tato práce se zabývá účinky triclosanu a triclocarbanu na významný biomarker epigenetické toxicity, tedy na mezibuněčnou komunikaci zprostředkovanou mezerovými spoji.

Abstract:

Triclosan and triclocarban are synthetic antimicrobial agents widely used in personal care products, cleaning products, human and veterinary pharmaceuticals and in industry. As a result of their excessive use and incomplete removal in wastewater treatment entering these substances into the environment, where they are detected both in surface waters, sediments and soils, and in aquatic and terrestrial organisms, including human tissues. Although they are not acutely toxic, mutagenic or teratogenic in mammals, these substances can have adverse effects on other organisms, particularly when their long-term effects. Their mechanisms of action include disruption of hormonal regulation and signaling, which is one of the so-called endocrine disruptors. This thesis discusses the effects of triclosan and triclocarban on the important biomarker of epigenetic toxicity, i.e. intercellular communication mediated mezerovými connections.
