Characterization and preparation of chimeric lectins from opportunistic pathogens

Abstract:

Tato práce se zaměřuje na lektiny z patogenních mikroorganismů. První část práce se zabývá krystalizací a řešením struktury hypotetického lektinu z organizmu z Burkholedia cepacia komplexu. Byly nalezeny a optimalizovány krystalizační podmínky, naměřena difrakční data a byla určena prostorová grupa, základní buňka a její rozměry. Poznání struktury nového lektinu BP39L má velký význam z hlediska pochopení jeho funkce a případného podílení se na patogenitě mikroorganismu. V druhé časti práce, zabývající se přípravou chimerních lektinů, byla prozkoumána možnost přípravy chimerních lektinů pomoci denaturace a renaturace dvou lektinů. Byla navrhnuta tvorba chimerních lektinů prostřednictvím koexpresního plazmidu pETDuet 1. Při práci byla odhalena zajímavá fakta o denaturaci a renaturaci lektinů z mikroorganismů Burkholderia cenocepacia a Pseudomonas aeruginosa Byla vypracována nová metodika zkoumání změn lektinů vyvolaných denaturačními činidly a detergenty. V návaznosti na tuto práci mohou byt zkoumány možnosti přípravy a vzniku chimerních lektinů, které mohou mít velké praktické uplatnění. Kromě praktického využití by studie napověděla, zda mohou vznikat lektinové chiméry i v samotných mikroorganizmech.

Abstract:

This work focuses on lectins from opportunistic pathogens. One part of work focuses on the crystallization and structure determination of a hypothetical lectin from a bacterium from Burkholedria cepacia complex. The crystal growth conditions were determined and optimized. The difraction data were collected and the space group and cell dimension was determined. The determination of the crystal structure of this new hypothetical lectin may have importance in understanding its function and can reveal the involvement of this lectin in pathogenic processes of the bacterium. The second part of the project deals with the preparation of chimeric lectins. Two approaches of their preparation were tried; first one using the denaturation and renaturation of two different lectins and second one using the co-expression system pETDuet 1. During the work, interesting facts were discovered by denaturation and renaturation of lectins from Burkholderia cenocepacia and Pseudomonas aeruginosa. To study the changes of lectins induced by the denaturants and detergents, new methodological approaches were developed. Thanks to this work, furthest possibilities of chimeric lectin creation can be investigated. The oligomeric chimera can have a great practical application and can verify the possibility of chimeric lectin creation in the bacterium.
