Cross talk of epidermal cells mediating protection against UV irradiation

Abstract:

Ochranu organizmu před ultrafialovým zářením mají na starosti buňky epidermis a jejich vzájemnou spoluprací dochází k tvorbě pigmentu melaninu, který záření absorbuje. Melanin se tvoří v melanocytech a je ukládán v podobě částic zvaných melanosomy. Pro lepší ochranu je nezbytné, aby se melanin dostal z melanocytů do keratinocytů, které tvoří většinu buněk epidermis. To je umožněno přenosem melanoso-mů, který probíhá různými mechanizmy. Melanogeneze a transport melaninu jsou řízeny různými faktory produkovanými okolními buň-kami. Tyto faktory spouští v melanocytech signální dráhy vedoucí k expresi melanogenních enzymů a proteinů transportu. Tato práce je soustředěna na reakci kokultivačního modelu melanocytů (B16 F10 nebo IPC 298) a keratinocytů (HaCaT) na UV záření. Po ozáření byla pozorována a změřena zvýšená úroveň melanogeneze. Nepřímo byl také detekován transfer melanosomů do keratinocytů. Nakonec byla kvantifikována exprese a aktivita vybraných proteinů, které řídí mela-nogenezi a transport melaninu, především p38, MITF a Mreg.

Abstract:

Protection against UV irradiation is dependent on the cells of the epi-dermis and their orchestrated cooperation that results in production of melanin pigment, which absorbs radiation. Melanin is formed in melanocytes and is stored in the form of vesicular melanosomes. For better protection, it is essential that melanin enters the keratinocytes that make up most of the epidermis. This is made possible by the transfer of melanosomes, which could occur by several mechanisms. Melanogenesis and melanin transport are controlled by various fac-tors produced by surrounding cells. These factors trigger signaling pathways in melanocytes leading to the expression of melanogenic enzymes and transport proteins. This thesis is focused on the reaction of the coculture model of melanocytes (B16 F10 or IPC 298) and keratinocytes (HaCaT) to UVB irradiation. Increased melanogenesis was observed and measured after irradiation. Transfer of the melano-somes to the keratinocytes was also detected indirectly. Finally, the expression and activity of selected proteins that control melanogenesis and melanin transport, especially p38, MITF and Mreg, were quanti-fied.
