Landscape genetics of cervids in western Carpathians and adjacent regions

Abstract:

Rešeršní část této práce se zabývá zejména popisem historických a ekologických aspektů fylogeografie jelena evropského (Cervus elaphus) a srnce obecného (Capreolus capreolus). Cílem práce je vypracovat empirickou studii zaměřenou na krajinnou genetiku cervidů, popisující populační strukturu C. elaphus a C. capreolus v Západních Karpatech a interpretovat vliv charakteru krajiny na genetickou strukturu. Sběr tkání daných druhů probíhal s využitím "citizen science". Byla využita zvířata legálně odlovená (popř. sražená dopravním prostředkem, nebo stržená predátorem) na území ČR, Slovenska a Polska. K analýze vzorků byla použita kontrolní oblast mitochondriální DNA (mtDNA). Výsledky ukazují vysokou haplotypovou diverzitu jak u srnce obecného, tak u jelena evropského. Na studovaném území dochází ke styku západní a východní haploskupiny jelena. Obě haploskupiny se prolínají v Moravskoslezských Beskydech, Podbeskydské pahorkatině, ve Slezských Beskydech, Vizovické vrchovině, Javorníkách, Bielych Karpatech, Kysuckých Beskydech a Strážovských vrších. Z výsledků také vyplývá, že pro jelena evropského tvoří rychlostní silnice E75 (ze Svrčinovce přes Čadcu, Žilinu, Bytču, Povážskou Bystricu) a dálnice D1 (z Žiliny na Trnavu) největší bariéru v migraci mezi Slovenskem a Českou republikou. U dvou jedinců srnce obecného z lokalit Povážské Podolie a Podbeskydská pahorkatina byla zjištěna introgrese mtDNA srnce sibiřského (Capreolus pygargus). Na studovaném území převládala centrální haploskupina. Výsledky ukazují, že srnci na sledovaném území spadají do jedné populace.

Abstract:

The research part of this thesis deals maiinly with the description of historical and ecological factors in phylogeography of the European deer (Cervus elaphus) and the roe deer (Capreolus capreolus). The aim of the work is to perform an empirical study based on landscape genetics of cervids in Western Carpathians, to analyze the population structure of C. elaphus and C. capreolus within the regionand interpret the impact of landscape features on the genetic structure. Collection of tissues of these species was carried out utilizing the citizen science approach. Samples were obtained from legally hunted and roadkilled animals and at the kill sites of large carnivoresin the Czech Republic, Slovakia and Poland. Mitochondrial DNA control region was used to analyze the samples. The results show a high haplotype diversity in both the European roe deer and the European deer. There is contact between the western and eastern haplogue groups of red deer within the study area. Both haplogroups intersect in the Moravian-Silesian Beskydy Mountains, the Podbeskydské Hills, the Silesian Beskydy Mountains, the Vizovické Highlands, the Javorníky Mountains, the Biele Carpathian Mountains, the Kysucké Beskydy Mountain and the Strážovské Mountains. The results also show that the E75 expressway (from Svrčinovec through Čadca, Žilina, Bytča, Povážská Bystrica) and the D1 motorway (from Žilina to Trnava) is the biggest barrier to migration between Slovakia and the Czech Republic. Two specimens of roe deer from Považské Podolie and Podbeskydská pahorkatina was introgressed by mtDNA of siberian deer (Capreolus pygargus). The central clade prevailed in the studied area. The results show that the roe deer belongs to one population.
