Methods of determining the antimicrobial activity of substances

Abstract:

Diplomová práce je zaměřená na stanovení antimikrobiální aktivity látek. Teoretická část práce je věnována oboru nanotechnologie a charakteristice nanomateriálů. Dále jsou charakterizovány jílové minerály sloužící jako matrice pro navázání nanočástic. Nejrozsáhlejší část uvádí průnik nanočástic do buněk, antimikrobiální aktivitu nanomateriálů a biologickou funkci nanočástic stříbra a zlata. V neposlední řadě je také zmíněno možné potencionální riziko nanočástic. Poslední kapitola teoretické části popisuje metody, kterými lze stanovit antimikrobiální aktivitu nanomateriálů. V experimentální části byla testována antibakteriální aktivita vybraných nanočástic na indikátorových bakteriích Escherichia coli, Enterococcus faecalis pomocí agarové diluční a mikrodiluční metody. Cílem experimentální části bylo stanovit minimální inhibiční koncentraci (MIC) a vyhodnotit získané výsledky.

Abstract:

The thesis is focused on the determination of the antimicrobial substances activity. The theoretical part is devoted to the field of nanotechnology and the characteristics of nanomaterials. Then there are characterized the clay minerals used as a matrix for the gaining of nanoparticles. The most extensive section provides an intersection of nanoparticles into cells, antimicrobial activity of nanomaterials and biological function of nanoparticles of silver and gold. Last but not least it is also mentioned the possible potential risk of nanoparticles. The last chapter of the theoretical part describes the methods by which one can determine the antimicrobial activity of nanomaterials. In the experimental part, there was tested the antibacterial activity of selected nanoparticles on indicator bacteria Escherichia coli, Enterococcus faecalis by using Agar dilution and micro-dilution methods. The aim of the experimental part was to determine the minimum inhibitory concentration (MIC) and to evaluate the results obtained then.
