Possibility of chitin and chitosan determination for quantification of fungal biomass

Abstract:

A number of fungi are known to be responsible for the production of usefull secondary metabolites with pharmaceutical use as well as those which are considered to be toxical.In the quality control of industrial scale food production analytical methods which are mainly focused on determination of biomarkers for fungal growth/biomass, and thus fungal spoilage, two such compounds are ergosterol and chitin. Chitin is a very well organised copolymer of 2-acetamido-2-deoxy- -D-glukosis, the main unit, and of 2-amino-2-deoxy- -D-glukosis,linked via the -(1-4) glucosidic bound. Chitin occurs as highly ordered microfibrils in many species in a variety of arrangements, from diatom spines to cell wall components of many fungi and yeast. Chitin is widely distributed in fungi, occuring in Basidiomycetes, Ascomycetes and Phycomycetes, where it is a component of the cell walls and structural membranes of mycelia and spores. Chitin serves as a fibrous strengthening element responsible for cell wall rigidity. When the degree of deacetylation of chitin reaches about 50% (depending on the origin of the polymer), it becomes soluble in aqueous acidic media and is called chitosan. A wide variety of medical applications for chitin and chitin derivatives have been reported over the past three decades. Various methods for chitin´s and chitosan´s determination are classified into three groups: spectroscopical, conventional and destructive.

Abstract:

Mnoho hub je zodpovědných za produkci velmi užitečných sekundárních metabolitů, jež mají farmaceutická využití, stejně tak jako ty, které jsou považovány za toxické. Pro stanovení kontaminace potravin plísněmi se v průmyslu využívají analytické metody, které jsou zaměřeny hlavně na stanovení biomarkerů biomasy a růstu plísní, dvěma takovými sloučeninami jsou ergosterol a chitin. Chitin je velmi dobře uspořádaný kopolymer, složený z 2-acetamido-2-deoxy- -D-glukózy, jež představuje hlavní složku, a z 2-amino-2-deoxy- -D-glukózy, spojených -(1-4) glukosidickou vazbou. Chitin se vyskytuje ve formě vysoce organizovaných mikrofibril v mnoha druzích, v pestré paletě uspořádání, od hřebenové až po součásti buněčných stěn hub a kvasinek. Chitin je v plísních hodně rozšířený, vyskytuje se u Basidiomycet, Ascomycet a Phycomycet, kde je součástí buněčné stěny a strukturní membrány mycelia a spór. Chitin slouží jako vláknité zesílení zodpovědné za pevnost buněčné stěny. Chitosanem nazýváme chitin, jehož stupeň deacetylace dosáhne asi 50 %, záleží však na původu polymeru, a jenž se stává rozpustným v kyselém vodném prostředí. V posledních třech desetiletích bylo hodně diskutováno využití chitinu a chitinových derivátů v různých oborech medicíny. Existují různé metody stanovení chitinu a chitiosanu, které jsou klasifikovány do tří skupin: spektroskopické, konvenční a destruktivní.
