Pseudomonas putida - growth and production of metalothioneins in presence of heavy metals

Abstract:

Znečištění životního prostředí ionty těžkých kovů je většinou důsledek antropogenní činnosti (těžební průmysl, hutnický průmysl, zemědělství). Na rozdíl od organických kontaminant, které mohou být přeměněny na látky méně toxické či netoxické, těžké kovy v přírodním prostředí perzistují. Často dochází k jejich akumulaci v nejrůznějších úrovních potravinového řetězce. Všechny organismy včetně bakterií si vytvořily mechanismy rezistence vůči těžkým kovům. Bakterie využívají mnoho rozdílných mechanismů obrany, příkladem je aktivní transport těžkých kovů z cytoplazmy do vnějšího prostředí (ATPázy, chemiosmotické membránové pumpy), extracelulární precipitace, intracelulární akumulace, volatizace či tvorba proteinů vázajících těžké kovy. Cílem práce bylo stanovení vlivu těžkých kovů (Cu2+, Co2+, Ni2+, Zn2+, Cd2+) na růst Pseudomonas putida CCM 4307. Tento mikroorganismus je integrální součástí konsorcia Biorem RC1 společně s Geotrichum candidum a Yarrowia lipolitica, které se využívá pro degradaci ropy ve znečištěném prostředí. Výsledky ukazují významné ovlivnění růstu a snížení hustoty buněčné suspenze s rostoucí koncentrací těžkých kovů v kultivačním prostředí a prodlužující se dobou kultivace. Druhá část této práce byla zaměřena na expresi metalothioneinům (MT) podobných proteinů studovaným kmenem kultivovaným v prostředí Cu2+, Co2+, Ni2+, Zn2+ a Cd2+. Největší množství tohoto proteinu bylo detekováno, jestliže byly buňky Pseudomonas putida CCM 4307 kultivovány v přítomnosti zinku.

Abstract:

Heavy metals are ubiquitous and persistent environmental pollutants that are introduced into the environment through anthropogenic activities (mining, smelting and agriculture). In contrast to organic pollutants, the contamination by heavy metals causes a serious problem because these metals cannot be naturally degraded and they accumulate in different parts of the food chain. All organisms including bacteria developed resistance mechanisms to toxic metals. Bacteria use many different systems of heavy metals defence, such as active transport of metals from cytoplasm (ATPases, chemiosmotic membrane pumps), extracellular precipitation of heavy metals, intracellular precipitation, passive diffusion of metal from cells and synthesis of binding proteins (methalothioneins and methalothioneins-like proteins). The first aim of this work was to analyze of effect of heavy metals (Cu2+, Co2+, Ni2+, Zn2+ and Cd2+) on the growth of Pseudomonas putida CCM 4307. This microorganism is integral component of consortium Biorem RC1 (together Geotrichum candidum and Yarrowia lipolitica) that is used for degradation of oil derivates including polyaromates. Results showed significant decrease of growth and density of cellular suspense in response to increasing concentration of heavy metals in cultural medium and elongating time of cultivation. The second aim of this work was to analyze effect of Cu2+, Co2+, Ni2+, Zn2+ and Cd2+ on the biosynthesize of metallothionein-like protein. Metallothionein are small, low-molecular weight, cystein-rich proteins. Bacterial metallothioneins were only identified in some cyanobacteria and pseudomonads. Results showed that the biggest amount of MT-like protein was detected when the studied microorganism was cultivated in presence of zinc.
