Vliv auxinových herbicidů na rovnováhu endogenních hladin IAA a jejích metabolitů u Hordeum vulgare a Arabidopsis thaliana

Effect of auxin herbicides on the homeostasis of endogenous levels of IAA and its metabolites in Hordeum vulgare and Arabidopsis thaliana

Abstract:

The effect of synthetic auxin herbicides (SAHs) on the regulation of endogenous auxin homeostasis in plants remains unclear. The aim of this thesis to reveal how endogenous metabolic pathways respond when monocot (Hordeum, SAH tolerant) and dicot (Arabidopsis, SAH susceptible) plants are exposed to exogenous IAA or auxin herbicides (2,4-D, dicamba, picloram and quinclorac). Sets of 6 Arabidopsis target genes (CYP79B2, CYP71A13, TAR2 and YUC2/4/6) and 7 barley genes (TSB, AMI, TAA/TAR, YUC, GH3.3, DAO and UGT74D1) were chosen and their relative expression was tested using RT-qPCR. To have further insight on regulation of IAA homeostasis, level of IAA precursors, conjugates and degradation products (ANT, TRP, TAM, IAOx, IAN, IAM, IPyA, IAA, IAA-Asp, IAA-Glu, oxIAA, oxIAA-Glc and IAA-Glc) were analyzed using method of solid-phase extraction and LC-MS/MS. The effect of auxin and SAHs on barley was obvious, especially decreasing of endogenous IAA levels after dicamba, picloram and quinclorac treatment, alternations in IAA precursor levels TAM and IPyA, and increases of auxin conjugates and degradation products correlated with increased expression of gene coding enzyme involved in IAA conjugation (GH3.3). The study in expression profiles in Arabidopsis reveal upregulation of the enzymes involve in IAA synthetics pathway typical for Brassicaceae, and increased expression of TAR2 after all treatments. The effect of IAA and auxin herbicides on Arabidopsis and barley showed surprisingly similar trends. These results may contribute to the understanding of how SAHs effect dicot and monocot plants, respectively on weeds and crops, differently and how is resistance to the SAHs developed.

Abstract:

Účinek syntetických auxinových herbicidů (SAH) na regulaci endogenní homeostázy auxinu zůstává u rostlin nejasný. Cílem této diplomové práce je odhalit reakce endogenních metabolických drah u jednoděložných (Hordeum, tolerantní vůči SAH) a dvouděložných (Arabidopsis, citlivé na SAH) rostlin vystavených exogennímu IAA nebo auxinovým herbicidům (2,4-D, dicamba, picloram a quinclorac). Byly vybrány sady 6 cílových genů Arabidopsis (CYP79B2, CYP71A13, TAR2 and YUC2/4/6) a 7 cílových genů ječmene (TSB, AMI, TAA/TAR, YUC, GH3.3, DAO and UGT74D1) jejichž relativní exprese byla testována pomocí RT-qPCR. Pro zisk dalších informací o udržování IAA homeostázy, hladiny IAA, jejích prekurzorů, konjugátů a produktů degradace (ANT, TRP, TAM, IAOx, IAN, IAM, IPyA, IAA, IAA-Asp, IAA-Glu, oxIAA, oxIAA-Glc a IAA-Glc) byly analyzovány pomocí metody extrakce na pevné fázi a LC-MS/MS. Účinek IAA a SAH na ječmen byl zřejmý zejména snížením endogenní hladiny IAA po ošetření herbicidy dicamba, picloram a quinclorac, změnou hladin IAA prekurzorů TAM a IPyA, zvýšením koncentrace IAA konjugátů a produktů degradace korelující se zvýšenou expresí genu kódujícího enzym zapojený do konjugace IAA (GH3.3). Studie expresních profilů Arabidopsis odhalila nadregulaci enzymů zapojených v IAOx syntetické dráze IAA typické pro Brassicaceae a také zvýšenou expresi TAR2 po všech ošetřeních. Účinek IAA a auxinových herbicidů na Arabidopsis a ječmen vykazoval překvapivě podobné trendy. Tyto výsledky mohou přispět k pochopení různých účinků SAH na dvouděložné a jednoděložné rostliny, respektive na plevele a plodiny, a jak se vyvíjí odolnost vůči SAH.
