Biologically active compounds of some parasitic plants from the Orobanchaceae family

Abstract:

Předložená disertační práce je zaměřena na botanickou a fytochemickou charakteristiku parazitických rostlin čeledi Orobanchaceae (zárazovité). Snahou bylo zejména rozšířit stávající informace o chemickém složení parazitických rostlin a pokusit se identifikovat u nich dosud nepopsané obsahové látky. K fytochemické analýze bylo vybráno celkem pět parazitických rostlin z uvedené čeledi. Největší pozornost byla věnována holoparazitickému druhu Lathraea squamaria L., včetně jeho dvou poddruhů. Pro porovnání chemického složení byly zvoleny čtyři rostliny poloparazitické, a to Euphrasia rostkoviana Hayne, Melampyrum nemorosum L., Melampyrum sylvaticum L. a Rhinanthus minor L. Zaměřili jsme se zejména na sloučeniny iridoidního a fenolického charakteru. Připravené extrakty z jednotlivých rostlinných částí byly analyzovány pomocí HPLC. Pro identifikaci obsahových látek v extraktech jsme využívali dostupné spektrální metody (UV, IR, MS, NMR). Izolace vybraných obsahových látek byla prováděna pomocí preparativní kapalinové chromatografie. U všech zkoumaných druhů jsme potvrdili přítomnost iridoidního glykosidu aukubinu, který má pro tyto rostliny chemotaxonomický význam. U L. squamaria jsme ve všech částech rostliny zaznamenali značné množství kyseliny benzoové, která u této rostliny nebyla dosud v literatuře uvedena, v menší míře je přítomna také u tří zkoumaných poloparazitických druhů, vyjma světlíku. Dalším novým poznatkem je přítomnost fenylpropanoidního glykosidu akteosidu a jeho isomeru isoakteosidu u obou poddruhů holoparazitického podbílku. Tyto látky jsme zaznamenali také u dvou poloparazitických zástupců, E. rostkoviana a R. minor. V případě Lathraea squamaria subsp. squamaria L. se nám podařilo získat větší množství rostlinného materiálu, proto bylo možné připravit dostatečný objem extraktu vhodného pro izolaci obsahových látek. Izolovali jsme a identifikovali fenylpropanoidní glykosidy akteosid a isoakteosid a iridoidní sloučeninu scyphiphin B1. Žádná z těchto látek dosud nebyla u tohoto druhu v literatuře popsána.

Abstract:

The presented thesis is focused on botanical and phytochemical characteristics of parasitic plants of the Orobanchaceae family to enlarge a knowledge of the chemical composition of parasitic plants and try to identify their constituents that have not been determined yet. Five parasitic plants of the mentioned family were chosen for the phytochemical analysis. The main subject of investigation was holoparasitic species Lathraea squamaria L., including both of its subspecies. Four semiparasitic plant species, Euphrasia rostkoviana Hayne, Melampyrum nemorosum L., Melampyrum sylvaticum L. and Rhinanthus minor L. were chosen for comparison of their chemical composition. We focused especially on the compounds of iridoid and phenolic structures. Prepared extracts of individual plant parts were analyzed by means of HPLC. The determination of content substances was carried out using available spectral methods (UV, IR MS, NMR). Isolation of chosen compounds was performed using preparative liquid chromatography. The presence of the iridoid glycoside aucubin, which has chemotaxonomical significance for these types of plants, was confirmed in all investigated parasitic species. Free benzoic acid was identified in all plant parts of L. squamaria for the first time, in smaller amount it was found out in extracts of three poloparasitic plants, except for eyebright (Euphrasia). Another new finding is the presence of phenylpropanoid glycoside acteoside and its isomeric isoacteoside in both subspecies of holoparasitic toothwort. These compounds were determined also in two semiparasitic species E. rostkoviana and R. minor. In case of Lathraea squamaria subsp. squamaria L. we managed to obtain a larger amount of plant material, therefore it was possible to prepare a sufficient volume of extract suitable for the isolation of the chosen constituents. We isolated and identified the phenylpropanoid glycosides acteoside and isoacteoside and the non-glycosidic iridoid compound scyphiphin B1. None of these substances have been determined in this species to date.
