Tablets coated by 3D printing and alcohol-induced dose dumping effect

Abstract:

Cílem této experimentální bakalářské práce bylo prostudovat možnosti 3D tisku pro potlačení "alcohol-induced dose dumping" efektu u matricových tablet s účinnou látkou tramadol hydrochlorid. Vybrané formulace (hydrofilní, lipofilní a směsná) matricových tablet byly připraveny metodou přímého lisování. U připravených tablet byly provedeny disoluční testy v kyselém a hydro-alkoholickém médiu (40 obj. % alkoholu) dle Českého lékopisu a vyhodnocen vliv alkoholu na uvolňování léčiva. S cílem eliminace vlivu alkoholu v disolučním médiu byla připravena další série tablet a tyto tablety byly potaženy metodou 3D tisku. Poté byl proveden disoluční test v hydro-alkoholickém médiu. Potahovým materiálem pro metodu 3D tisku byl polymer PVA. Disoluční chování potahovaných tablet v disolučním médiu s obsahem 40 obj. % alkoholu bylo srovnáno s profilem nepotahovaných tablet. Uvolněné množství tramadol hydrochloridu bylo stanovováno pomocí UV/VIS spektrometrie. Získaná data byla kvantitativně vyhodnocena pomocí vhodných matematických modelů (model Weibull a kinetický model I. řádu). Na základě kinetických parametrů získaných nelineární regresní analýzou disolučních profilů byl vyhodnocen vliv potahu tablety na uvolňování léčiva v hydro- alkoholickém médiu a potlačení "alcohol- induced dose dumping" efektu.

Abstract:

The goal of this experimental bachelor's thesis was to study the possibilities of 3D printing in suppression the "alcohol-induced dose dumping" effect in matrix tablets with tramadol hydrochloride as active substance. Selected formulations (hydrophilic, lipophilic and mixed) of matrix tablets were prepared by direct compression. The prepared tablets were subjected to dissolution tests in acidic and hydro-alcoholic medium (40% alcohol) according to the Czech Pharmacopoeia and then evaluated by the effect of alcohol on drug release. In order to eliminate the effect of alcohol in the dissolution medium, another series of tablets was prepared and these tablets were coated by 3D printing. Then a dissolution test was done in a hydro-alcoholic medium. The coating material for the 3D printing method was PVA polymer. Finally, the dissolution behaviour of both, the coated tablets in a dissolution medium containing 40% alcohol and the profile of the uncoated tablets were compared. The amount of released tramadol hydrochloride was determined under UV / VIS spectrometry. The received data were quantitatively evaluated by using suitable mathematical models (Weibull model and first-order kinetic model). Based on the kinetic parameters obtained by nonlinear regression analysis of dissolution profiles, the effect of the tablet coating on drug release in a hydro-alcoholic medium and suppression of the "alcohol-induced dose dumping" effect was evaluated.
