Toxic effects and biochemical changes after inhalation exposure to nanoparticles

Abstract:

Nanomateriály mají řadu využití v oblasti vědy, techniky a medicíny, ale je málo informací ohledně jejich potenciálního působení na organismy včetně člověka a i proto jsou v současné době často diskutovaným tématem. První část práce je zaměřena na vlastnosti nanočástic, jejich rozdělení a zdroje. Druhá část práce se zabývá toxickými vlastnostmi nanočástic a jejich vlivem od buněčné úrovně až po zdraví člověka. V této části práce je diskutován oxidativní stres, který je považován za důležitý mechanismus působení nanočástic a biochemické markery k jeho sledování. Praktická část práce je založená na studiu ovlivnění vybraných biomarkerů oxidativního stresu po inhalační expozici modelového organismu Mus Musculus nanočásticemi oxidů manganu a oxidu kademnatého. Při akutní expozici byly myši vystaveny nanočásticím po dobu 4, 12, 24 a 72h, chronická expozice trvala 11, 13, 15 a 17 týdnů. Ve tkáních jater, mozků a plic byly stanoveny aktivity enzymu glutation-S-transferáza, úroveň lipidní peroxidace a hladiny redukovaného a oxidovaného glutationu. Z naměřených výsledků vyplývá, že studované nanočástice mají potenciál způsobovat oxidativní stres. Dlouhodobější expozice může vést ke zvýšené peroxidaci lipidů a to zejména ve tkáních jater a mozků. Zjištěné účinky jsou v práci diskutovány s dalšími studiemi, které se zabývají inhalační expozicí nanočástic.

Abstract:

Nanomaterials are used in science, technology and medicine but there is still limited knowledge on their potential effects on organisms; therefore, they tend to be a widely discussed topic in these days. The first part of the thesis focuses on the properties, types and sources of nanoparticles. The second part of the thesis deals with toxic properties of nanoparticles and their impact from cellular level to human´s health. This part of the thesis also discusses oxidative stress as an important mechanism involved in nanoparticles´ effects on organisms and biochemical markers for its evaluation. The practical part of the thesis is based on the study of selected oxidative stress biomarkers after inhalation exposure of model organism Mus Musculus to manganese and cadmium nanoparticles. During an acute exposure mice were exposed to nanoparticles for 4, 12, 24 and 72h, chronic exposure lasted for 11, 13, 15 and 17 weeks. Activities of glutathione-S-transferase, lipid peroxidation and levels of oxidized and reduced glutathione were measured in liver, brain and lung tissues. Results imply that nanoparticles have potential to cause oxidative stress. Longer exposure can lead to peroxidation of lipids especially in liver and brain tissues. Observed effects are discussed with other studies concerned with inhalation exposure of nanoparticles.
