Does geographic location affect plant leaf phenotype? A case study of the Euro-Siberian Ranunculus auricomus species complex using geometric morphometrics and bioclimatic data

Abstract:

Rozsáhlá fenotypová plasticita rostlinných orgánů umožňuje adaptaci rostlin na měnící se podmínky životního prostředí. Nejvýraznější fenotypovou variabilitu vykazují listy. Variabilita listových forem byla studována zejména u dřevin, menší pozornosti jí bylo věnováno u bylin. Druhový komplex Ranunculus auricomus agg. je intenzivně studován jako model evoluce fylogeneticky mladých rostlinných druhů. Ačkoliv tvar listů sloužil jako významné vodítko v taxonomii komplexu, bylo zřejmé, že jeho velká variabilita znemožňuje korektní klasifikaci jednotlivých druhů. Fenotypová plasticita bazálních listů jedinců komplexu R. auricomus může být ovlivněna faktory prostředí a určité fenotypy listů se mohou vyskytovat v určitých biotopech. V této diplomové práci jsem testoval hypotézu, že fenotypy bazálních listů jedinců komplexu R. auricomus se vyskytují v závislosti na konkrétních podmínkách prostředí. Tuto hypotézu jsem testoval na základě 512 herbářových položek pokrývající celou oblast západní Eurasie. Pro každou položku jsem získal doprovodná ekologická data: bioklimatické proměnné a charakter geologického podloží lokality. Cílem bylo odpovědět na tyto otázky: (1) Jaké listové fenotypy komplexu R. auricomus se vyskytují východně od dosud studovaných oblastí, tzn. ve východní Evropě a v západní části Ruské federace? (2) Existuje souvislost mezi typem klimatu (oceánický - kontinentální) a listovými fenotypy populací komplexu R. auricomus? Lze vysledovat rozdíly mezi populacemi na západě a na východě Evropy? (3) Které ekologické faktory lze označit za prediktory distribuce listových fenotypů v komplexu R. auricomus napříč západní částí Eurasie? Analyzoval jsem 1002 bazálních listů jedinců R. auricomus pomocí metod landmarkové geometrické morfometriky. Souvislost mezi tvary bazálních listů a faktory prostředí jsem testoval pomocí vybraných modelů distribuce druhů (logistická regrese, model maximální entropie (MaxEnt)). Geometrická morfometrika poukázala na hlavní morfologický gradient v tvaroprostoru bazálních listů: tvarový trend od téměř nečleněných bazálních listů k členěným listům s velmi hlubokými zářezy dosahujícími až k hlavní žilce (56 % celkové tvarové variability). Druhý nejvýznamnější tvarový trend byl charakterizován zvětšujícím se úhlem úkrojků při bázi listové čepele (20 % celkové tvarové variability). Regresní modely ukázaly, že zatímco se nečleněné fenotypy bazálních listů vyskytují častěji v oblasti s kontinentálním typem klimatu, výskyt členěných fenotypů je vázán především na oblast s oceánickým typem klimatu. Jinými slovy, gradient členitosti listů významně souvisel s průměrnou teplotou nejteplejšího čtvrtletí (BIO9) a teplotní sezónností (BIO4). Tvarový trend, který sleduje oceánicko-kontinentální klimatický gradient, může mít ekofyziologické opodstatnění související s efektivní termoregulací rostliny v období s nejnižším úhrnem srážek. Lze konstatovat, že členěné a nečleněné fenotypy listů jedinců R. auricomus nejsou rozšířeny v západní Eurasii rovnoměrně, ale že jejich rozšíření sleduje bioklimatické veličiny spojené s kontinentálně-oceánickým gradientem.

Abstract:

Extensive phenotypic plasticity of plant organs allows plants to adapt to changing environmental conditions. Among plant organs, leaves show the most notable phenotypic variability. While the variation in leaf forms was studied mainly in woody plants, less attention was paid to herbs, except for the complex Ranunculus auricomus agg. This complex of closely related species was established as a model system for studying young (Quaternary) plant speciation. Although the shape of the basal leaves served as an important character in the taxonomy of the complex, it was clear that its considerable variation made it impossible to correctly classify species. The phenotypic plasticity of the basal leaves might be influenced by environmental factors, and specific leaf phenotypes might be preferentially expressed under particular environmental conditions. In this diploma thesis, I tested the hypothesis, that environmental conditions predict the geographic distribution of basal leaf phenotypes in the R. auricomus complex. I tested this hypothesis based on 512 herbarium specimens covering major parts of Western Eurasia. For each herbarium specimen, I collected images of basal leaves along with accompanying ecological metadata characterizing the sampling spots, including bioclimatic variables and geology. I aimed at answering the following three questions: (1) What leaf phenotypes of the R. auricomus complex occur east to the so far studied areas, eg. in Eastern Europe and Russian Federation? (2) Is there any relationship between climate (oceanic - continental) and leaf phenotypes of R. auricomus populations and can differences be traced between populations in western and eastern Europe? (3) Which ecological factors can be identified as predictors of the distribution of leaf phenotypes in the R. auricomus complex across the western part of Eurasia? I analyzed 1002 basal leaves of R. auricomus individuals using landmark geometric morphometrics. I tested the relationship between the leaf shape and environmental factors at sampling sites using selected species distribution models (logistic regression, maximum entropy model (MaxEnt)). Geometric morphometrics pointed to the main morphological gradient in the basal leaf morphospace: the shape trend from almost non-dissected leaves to dissected leaves with deep incisions reaching to the main leaf vein (56 % of the total shape variability). The second most significant shape trend was characterized by an increasing angle of the segments at the base of the leaf blade (20 % of the total shape variability). Regression models have shown that while non-dissected basal leaf phenotypes occur more frequently in areas under more continental conditions, the higher occurrence of fragmented phenotypes is related to areas with a more oceanic climate. In other words, the gradient of leaf dissection was significantly related to the mean temperature of driest quarter (BIO9) and temperature seasonality (BIO4). The shape trend, which follows the oceanic-continental climate gradient, might have an ecophysiological explanation related to the effective thermoregulation of the plant in the period with the lowest total precipitation. It can be concluded that the non-dissected and dissected leaf phenotypes of R. auricomus individuals are not evenly distributed in western Eurasia, but that their distribution follows bioclimatic conditions associated with the continental-oceanic gradient.
