Modulation of redox system and sulfur metabolism by cytokinin in Arabidopsis thaliana

Abstract:

Modulace řady buněčných procesů cytokininy je spojena s redoxními změnami. Antioxidační a redoxní systémy spojuje tripeptid glutathion, který tak významně určuje redoxní stav buňky. Právě obsah glutathionu byl po působení cytokininů snížen, zatímco snížení hladiny endogenních cytokininů vedlo ke zvýšení obsahu glutathionu. Tyto změny obsahu glutathionu nebyly doprovázené rovnocennou změnou obsahu oxidovaného glutathionu, a mohou tedy být spojeny se změnou redoxního potenciálu glutathionu. Glutathion představuje zásobní formu redukované síry, a proto byl dále sledován účinek cytokininů na metabolizmus síry. Působení cytokininů vedlo k nadregulaci transkripčních markerů odpovědi na nedostatek síry. Účinek cytokininů nevyžadoval klíčový transkripční faktor odpovědi na nedostatek síry SULFUR LIMITATION 1 a snížení obsahu glutathionu bylo nezávislé na aktivitě γ-glutamylcyklotransferázy GGCT2;1, která katalyzuje degradaci glutathionu a je robustně nadregulována cytokininy. Biologická relevance kolísání hladin glutathionu při změnách obsahu cytokininu byla prokázána modulací tolerance ke kademnatým iontům a organickým agens, které depletují glutathion. Negativní účinek cytokininů na obsah glutathionu a nadregulace transkripčních markerů nedostatku síry ukázaly na význam cytokininů v regulaci výživy sírou a dále naznačují možné mechanizmy, kterými cytokininy mohou interagovat s komplexní sítí buněčné signalizace.

Abstract:

Cytokinin action is coupled with redox-related evens. Tripeptide glutathione is a key compound linking antioxidant and redox systems and, thus, glutathione constitutes a significant determinant of the redox state of the cell. Cytokinin level and signaling negatively correlated with glutathione content whereas cytokinin treatment was coupled with stable or even elevated contents of oxidized glutathione supporting an in-dication for a shift in the redox state of the cell in response to cytokinin. Cytokinin-induced glutathione depletion apparently originates from the metabolic glutathione utilization or degradation, however, it is independent of glutathione degrading en-zyme, GAMMA-GLUTAMYL CYCLOTRANSFERASE 2;1, expression of which was robustly upregulated by cytokinin. A physiological impact of glutathione depletion was underpinned by increased cadmium sensitivity of cytokinin-treated Arabidopsis. In accordance, cytokinin-deficient plants displayed improved root growth upon expo-sure to glutathione-depleting chemicals that was attributed to a higher capacity to maintain glutathione levels. Exogenous cytokinin triggered the sulfur starvation-like gene expression accompanied by a decrease of sulfate along with glutathione content. This process was uncoupled from the activity of the major transcriptional regulator of sulfate starvation signaling SULFUR LIMITATION 1. The links between cytokinin and sulfur starvation-responsive gene expression and glutathione levels suggest a key role for cytokinin in sulfur nutrition. Moreover, the involvement of glutathione in many cellular processes suggests that altered glutathione might function in crosstalk between cytokinin and hormonal and environmental signaling pathways.
