In vitro assessment of intra- and intercellular signalling in response to environmentally occurring substances

Abstract:

Hlavním cílem disertace byla charakterizace in vitro modelu lidské dýchací soustavy reprezentovaného imortalizovanou lidskou bronchiální buněčnou linií HBE1 ve smyslu vhodnosti a použitelnosti k hodnocení narušení mezi-/vnitro-buněčného signálování jako důsledku expozice látkám vyskytujícím se v prostředí. Prezentované výsledky se zaměřují převážně na dva aspekty buněčné komunikace, a to inhibici mezibuněčné komunikace zprostředkované mezerovými spoji (z angl. gap junctional intercellular communication; GJIC) a změny vnitrobuněčného signálování skrze mitogenem aktivované proteinkinázy (MAPK), a poskytují tak nový náhled na model inhalační expozice lidského dýchacího systému. Pro zhodnocení vlivů toxikantů, sinicového toxinu Microcystinu-LR (MCLR) a nízko-/středně-molekulárních polycyklických aromatických uhlovodíků, na vnitrobuněčné signálování, životaschopnost a GJIC byly použity zavedené metody (např. Western blotting, trojkombinace testů viability, scrape-loading/dye transfer assay; SLDT). Nedostatečná testovací kapacita tradičně používané metody SLDT představovala významné omezení, proto byla zavedena modernizovaná metoda s vysokou propustností umožňující automatické hodnocení několika parametrů současně. Použitelnost modernizované metody byla ověřena otestováním škály látek s různým mechanismem účinku. Dále byla také využita ke zhodnocení potenciálně chemopreventivních účinků C-geranylovaných flavonoidů izolovaných z listnatého stromu Paulownia tomentosa a jejich schopnosti obnovovat GJIC v linii nekomunikujících tumorigenních in vitro jaterních potkaních buněk.

Abstract:

The main established aim of the dissertation was to characterize the in vitro model of human respiratory system, represented by immortalized human bronchial epithelial HBE1 cells, in the sense of suitability for the determination of intra-/intercellular signalling disruption caused by the exposure to environmentally occurring substances. The herein presented results follow two main aspects of cellular communication, namely the inhibition of intercellular signalling carried through gap junctions (gap junctional intercellular communication; GJIC) and changes in intracellular signalling represented by the mitogen-activated protein kinases (MAPKs) pathways respectively and provide a new perspective into the in vitro inhalation-like exposure model of a human respiratory system. Well-established methods (e.g. Western blotting, cytotoxicity assays, scrape-loading/dye transfer assay; SLDT) were utilized to determine effects of cyanobacterial toxin Microcystin-LR and low-/middle-molecular weight polycyclic aromatic hydrocarbons (LMW/MMW PAHs) on intracellular signalling, viability and GJIC respectively. The throughput of traditional SLDT assay represented a significant bottleneck and lead to the introduction of a novel, high throughput-compatible SLDT allowing automated and simultaneous evaluation of multiple parameters. Later on, this novel method was evaluated to be suitable for assessment of multiple agents with different modes of action in different cell lines, including human bronchial epithelial cells. Furthermore, the method was utilized to assess potentially chemopreventive compounds, namely C-geranylated flavonoid substances isolated from a broadleaf tree Paulownia tomentosa, and their ability to attenuate GJIC-deficient in vitro tumorigenic rat liver epithelial cells.
