Oxidation and reduction of proteins on the electrically charged surfaces

Abstract:

Fast progress in proteomics and biomedicine requires new methods of protein analysis. Many various analytical techniques are used for characterization of proteins, and also for diagnostics purposes in biomedicine. Electrochemistry represents another interesting method in protein analysis. In recent decades, the electrochemistry of proteins was focused mainly on conjugated proteins containing redox-active residues (eg. metalloproteins) providing reversible voltammetric signals. This interesting bioelectrochemistry worked with a relatively small group of proteins, while thousands of proteins studied in the proteomics and important in biomedicine were beyond the reach. In our laboratory, new method was developed based on a combination of the ability of proteins to catalyze hydrogen evolution at mercury electrodes with the constant-current chronopotentiometry. The aim of the main part of this Thesis was to try to elucidate the role of different types of amino acid residues in the electrocatalysis. In this Dissertation Thesis, the results of electrochemical analysis of peptides and proteins at mercury and carbon electrodes are presented. This work includes three self contained studies. The first two studies are aimed at research of amino acid residues with positively charged side chains at mercury electrodes. Effect of individual amino acid residues were studied with model systems (polyamino acids and hexapeptides) using voltammetry and constant current chronopotentiometric analysis. Our work with hexapeptides shows that the effect of the protonation of amino acid side chain plays important role in the catalytic hydrogen evolution reaction. Our results with model system clarified the influence of the amino acid composition on the properties of peak H and contributed to a better understanding of its nature. The third work discuss electro-oxidation of the protein AGR2, which participates in mechanisms of cancer diseases. In our study, this protein and its mutant E60A, were for the first time detected using carbon electrodes at nanomolar concentrations. The study experimentally confirmed the oxidation of histidine residues in the proteins and showed influence of the accessibility and number of histidine residues on the oxidation peak of histidine. This work could make studies at the carbon electrodes more attractive in protein analysis, because in addition to the peak of tyrosine and tryptophan residues, the oxidation peak of histidine residues could be also followed.

Abstract:

Rychlý pokrok v biomedicíně a genomice vyžaduje nové metody analýzy proteinů. V dnešní době se používá velké množství analytických metod pro charakterizaci proteinů a také v biomedicíně pro diagnostické účely. Elektrochemie představuje další zajímavou metodu v proteinové analýze. V posledních desetiletích byla elektrochemie bílkovin soustředěna především na konjugované bílkoviny obsahující redoxně aktivní zbytky (např. metaloproteiny) poskytující reversibilní voltametrické signály. Tato zajímavá bioelektrochemie pracovala však s relativně malým počtem bílkovin, zatímco tisíce bílkovin studovaných v rámci proteomiky a významných v biomedicíně bylo mimo její dosah. V naši laboratoři byla navržena nová metoda založená na schopnosti bílkovin katalyzovat vylučování vodíku na rtuťových elektrodách v kombinaci s chronopotenciometrií s konstantním proudem. Cílem větší části této disertace bylo přispět k objasnění úlohy různých typů zbytků aminokyselin v studované elektrokatalýze. V předkládané práci jsou prezentovány výsledky elektrochemických analýz peptidů a proteinů na rtuťových a uhlíkových elektrodách. Práce zahrnuje tři ucelené studie. První dvě studie jsou zaměřeny na zkoumání aminokyselin s pozitivně nabitým postranním řetězcem na rtuťových elektrodách. Vliv jednotlivých aminokyselinových zbytků byl zkoumán nejdříve u polyaminokyselin a později u hexapeptidů pomocí voltametrických metod a chronopotenciometrické analýzy konstantním proudem. Práce s hexapeptidy ukazuje vliv protonizace postranní skupiny aminokyselin na katalytické vylučovaní vodíku. Naše výsledky přispěly k objasnění vlivu aminokyselinového složení na vlastnosti píku H a pomohly k lepšímu porozumění jeho podstaty. Třetí práce se zaobírá elektro-oxidací proteinu AGR2, který se účastní mechanizmů důležitých v onkologických onemocnění. V naší studii byl tento protein a jeho mutant E60A poprvé detegován pomocí uhlíkových elektrod v nanomolárních koncentracích. Práce se zabývá oxidací histidinových zbytků v této bílkovině a vlivem dostupnosti a počtu histidinových zbytků na oxidační pík histidinu. Tato práce by mohla přispět k zefektivnění studia na uhlíkových elektrodách, kde by kromě píku tyrosinových a tryptofanových zbytků mohl být sledován také oxidační pík histidinových zbytků.
