Capillary electrophoresis of quantum dots and their conjugates with proteins

Abstract:

Tématem této práce je především optimalizace metodiky syntézy, separace a detekce CdTe nanokrystalů a jejich konjugátů s biomolekulami pro účely fyziologického výzkumu a molekulární diagnostiky. Luminiscenční sondy byly připraveny konjugací luminiscenčních sloučenin s biomolekulou. Jako luminiscenční značky byly použity CdTe polovodičové nanokrystaly - kvantové tečky. Takto značené molekuly jsme detekovali pomocí kapilární zónové elektroforézy s laserem indukovanou fluorescenční detekcí. Cílem této práce je vývoj a optimalizace metody značení biologicky významných molekul kvantovými tečkami pomocí tzv. "zero-length crosslinkerů" pro jejich budoucí aplikaci ve funkční diagnostice apoptotických a nádorových buněk.

Abstract:

The main themes of this thesis are optimize the synthesis methodology, separation and detection of CdTe nanocrystals and their conjugates with biomolecules for the purpose of physiological research and molecular diagnostics. Luminescence probes were prepared by conjugation of luminescent compound with biomolecules. CdTe semiconductor nanocrystals - quantum dots were used as luminescent markers. Prepared conjugates were detected by capillary zone electrophoresis with laser induced fluorescence detection. The aim of this work is to develop and optimize methods for labeling biologically important molecules with quantum dots via "zero-length crosslinkers" for their future application in functional diagnostic of apoptotic and tumor cells.
