Electrochemical Detection Of Ergosterol As An Indicator Of Food Contamination By Fungi

Abstract:

Cílem práce bylo zavést a optimalizovat metodu elektrochemického stanovení ergosterolu. Teoretická část shrnuje informace o ergosterolu, jeho syntéze, využití a jeho souvislost s plísněmi, které mohou být nežádoucími kontaminanty především potravinových vzorků. Další kapitoly popisují možnosti stanovení ergosterolu. Vlastní experimentální část byla zaměřena na zavedení a optimalizaci podmínek detekce ergosterolu square-wave voltametrie. Byly testovány tři typy elektrod, vybráno vhodné reakční prostředí a stanoveny detekční limity. Metoda byla optimalizována s využitím standardu ergosterolu, v závěru byl testován vzorek směsi plísní. Ve výsledkové části jsou experimenty diskutovány.

Abstract:

The aim of the work was to introduce and optimize the method of electrochemical determination of ergosterol. The theoretical part summarizes information about ergosterol, its synthesis, use and its relationship with fungi, which can be undesirable contaminants, especially food samples. The next chapters describe the possibilities of determining ergosterol. The experimental part was focused on the introduction and optimization of condition for the detection of ergosterol square-wave voltammetry. Three types electrodes were tested, a suitable reaction medium was selected and detection limits were set. The method was optimized using the ergosterol standard, in the end a sample of the mold mixture was tested. In final part, the experiments are discussed.
