Micronucleus test and its use for detection of genotoxicants

Abstract:

Tato práce byla zaměřena na možnosti využití Mikronukleus testu pro detekci genotoxických agens. Řada látek, nacházejících se v životním či pracovním prostředí člověka, vykazují nebezpečné vlastnosti. Velkou pozornost si v poslední době vyžádaly látky karcinogenní, u kterých až v 85 % případů můžeme sledovat jejich mutagenní, potažmo genotoxický efekt. Jednou z nejlepších možností pro určení expozice osob karcinogenními chemickými látkami je biologický monitoring, proto je praktická část této práce věnována sledování frekvence mikrojader v cytoplazmě dvoujaderných buněk, které jsou odrazem projevu genotoxického působení na organismus. Pro posouzení byla využita skupina pracovníků, pracujících v chemickém závodě, kteří jsou ve svém pracovním procesu exponováni sloučeninám niklu a kadmia. Výsledky této mikroskopické analýzy byly statisticky porovnány s výsledky předchozího roku, kdy byl uvedenou metodou testován stejný soubor pracovníků.

Abstract:

The bachelor thesis was focused on usage of Micronucleus test for detection of genotoxic agents. Lots of substances, occuring in the human living or working environment, show their dangerous character. Great attention has recently called for carcinogenic substances, for which up to 85 % can be traced to their mutagenic and genotoxic effects. One of the best possibilities for determining the exposure of the carcinogenic chemical substances is biological monitoring. Practical part of this work is devoted to monitoring the frequency of micronuclei in the cytoplasm of binuclear cells, which reflect the expression of genotoxic effects on the organism. There was used to group of workers exposed to cadmium and nickel compounds. Results of microscopically analysis was compared with results of previous year, when the same workers was analysed by Micronucleus test.
