Mikroplasty ve vodním prostředí: srovnání metod separace mikroplastů z biomasy vodních bezobratlých

Microplastics in aquatic environment: comparison of methods for microplastics separation from aquatic invertebrates biomass

Abstract:

Kontaminace životního prostředí a zejména vodních ekosystémů mikroplasty je v současnosti velmi diskutované téma s celospolečenským dopadem. Pro posouzení míry kontaminace vodního prostředí mikroskopickými částicemi plastu včetně posouzení jejich dopadu na vodní biotu není k dispozici jednotná metodika. Neexistují tudíž postupy, které by standardizovaly odběr a úpravu vzorků vodních bezobratlých, kteří slouží celosvětově jako bioindikátoři kvality vodního prostředí, pro posouzení množství či typu mikroplastů v nich obsažených. Na základě studia dostupné literatury byly vybrány čtyři metody, kterými lze biomasu vodních bezobratlých rozložit a poté stanovit množství mikroplastů obsažených v jejich tělech. Jedná se o rozpouštění pomocí kyseliny dusičné, peroxidu vodíku, hydroxidu sodného a proteinázy K. Tyto metody byly prakticky testovány a ověřeny na třech modelových taxonech vodních bezobratlých ze čtyř lokalit v podélném gradientu řeky Svratky. Metody byly porovnány a díky celkově nadprůměrným počtům nalezených mikroplastů, ale také díky vizuálně úspěšnější degradaci biomasy byla pro budoucí práce jako ideální postup zvolena metoda separace pomocí peroxidu vodíku. Průměrně bylo v 1 mg sušiny nalezeno 2,53 kusů mikroplastů u rodu Baetis, 0,21 kusů mikroplastů u rodu Calopteryx a průměrně 1 mikroplast u rodu Hydropsyche. Ze všech tří vybraných taxonů se rod Hydropsyche jeví jako nejvhodnější modelový taxon pro tento typ výzkumu. Při porovnání počtu nalezených mikroplastů mezi jednotlivými modelovými lokalitami nebyl pozorován žádný trend zvyšování počtu mikroplastů od pramene řeky k ústí. Hlavním výstupem této bakalářské práce je „Návrh metodiky odběru, zpracování a vyhodnocení vzorků mikroplastů z makrozoobentosu tekoucích vod“.

Abstract:

Environmental contamination by microplastics and especially in aquatic ecosys-tems is currently a highly-discussed topic with notable impacts. There is no unified methodology for assessing the contamination of the aquatic environment by microscopic plastic particles, including the assessment of their impact on aquatic biota. Thus, there are no methods to standardize the sampling and treatment of aquatic invertebrates, which serve as bioindicators of aquatic quality worldwide, to assess the amount or type of microplastics they contain in their body mass. Based on the study of the available literature, four methods were selected by which the biomass of aquatic invertebrates can be decomposed and then the number of microplastics contained in their bodies determined. These are dissolution using nitric acid, hydrogen peroxide, sodium hydroxide and proteinase K. These methods were practically tested and verified on three model taxa of aquatic invertebrates from four localities in the longitudinal gradient of the Svratka river. The methods were compared, and due to the overall above average number of microplastics found, but also due to the visually more successful degradation of biomass, the method of separation using hydrogen peroxide was chosen as an ideal method for future work. On average, 2.53 microplastics in the genus Baetis, 0.21 microplastics in the genus Calopteryx and an average of 1 microplastic in the genus Hydropsyche were found in 1 mg of dry matter. Of all three selected taxa, the genus Hydropsyche appears to be the most suitable model taxon for this type of research. When comparing the number of microplastics found between the individual model localities, no trend of increasing the number of microplastics from the source of the river to the estuary was observed. The main output of this bachelor's thesis is "Proposal of a methodology for sampling, processing and evaluation of microplastic samples from benthic macroinvertebrates in running freshwater".
