Possibilities of using flow cytometry in laboratory diagnosis of sepsis

Abstract:

Teoretická část bakalářské práce se zabývá popisem průtokové cytometrie, typy analýz a jejím využitím v klinické diagnostice i mimo medicínu. Dále je zpracována část o sepsi a laboratorní diagnostice sepse. Praktická část bakalářské práce je zaměřena na stanovení exprese antigenu HLA-DR na monocytech a regulačních T-lymfocytů. Byla provedena statistická analýza závislostí vztahu vybraných biochemických parametrů C-reaktivní protein, prokalcitonin, interleukin-6 a presepsin a také hematologických parametrů leukocyty, hemoglobin, trombocyty a fibrinogen s parametry CD14/HLA-DR a regulační T-lymfocyty. Byla provedena korelační analýza pro porovnaní výše uvedených metod s CD14/HLA-DR a T regulačními lymfocyty. Parametr CD14/HLA-DR nejvíce koreloval s leukocyty, fibrinogenem a C-reaktivním proteinem. Regulační T-lymfocyty korelovaly nejlépe s fibrinogenem. Na základě statistické analýzy bylo zjištěno, že soubory pacientů se sepsí a bez sepse u metody CD14/HLA-DR a Tregs mezi sebou mají statisticky odlišné výsledky.

Abstract:

The theoretical part of the bachelor thesis deals with the description of flow cytometry, types of analyses and their use in clinical diagnostics and outside of medicine. Next, there is a processed part on sepsis and laboratory diagnostics of sepsis. The practical part of the bachelor thesis is focused on the determination of antigen´s expression HLA-DR on monocytes and regulatory T-lymfocytes. There was performed a statistical analysis of the relationship between selected biochemical parameters C-reactive protein, procalcitonin, interleukin-6 and presepsin and also hematological parameters leukocytes, hemoglobin, platelets and fibrinogen with parameters CD14/HLA-DR and regulatory T-lymfocytes. Correlation analysis was performed for compare the above methods with CD14/HLA-DR and T regulatory lymfocytes. The parameter CD14/HLA-DR was the most correlated with leukocytes, fibrinogen and C-reactive protein. Regulatory T-lymfocytes correlated best with fibrinogen. On base of statistical analysis was found, that the files of patients with sepsis and without sepsis in the method of CD14/HLA-DR and Tregs between themselves have statistically different results.
