Epigenetic mechanisms regulating activity of the plant telomerase

Abstract:

Regulace exprese a aktivity telomerázy probíhá na několika úrovních. Jednou z nich je i epigenetická regulace genové exprese, která zahrnuje methylaci cytosinů, modifikace histonů, působení enzymů remodelujících chromatin a vliv nekódujících RNA. Tato práce v teoretické části shrnuje epigenetické mechanismy regulace genů, se zaměřením na methylaci cytosinů; dále se věnuje regulaci proteinové podjednotky telomerázy a vzhledem k části praktické i telomerám a jejich epigenetickým modifikacím. V praktické části se práce věnuje analýze aktivity telomerázy v pletivech A. thaliana, transkripce genu pro katalytickou podjednotku telomerázy a analýze epigenetických modifikací chromatinu tohoto genu. Srovnány jsou rostliny divokého typu s rostlinami nesoucími T-DNA inzerci v genech DDM1 a MET1, jejímž výsledkem je snížení celkové methylace cytosinů v genomu A. thaliana. Dále je řešena stabilita telomer a methylace cytosinů v telomerických sekvencích v rostlinách s hypomethylovaným genomem. V části Diskuze jsou výsledky experimentů probrány s ohledem na aktuální stav výzkumu epigenetické regulace telomerázy a telomer v různých modelových organismech.

Abstract:

Regulation of telomerase expression is the multilevel proces and epigenetic mechanisms including methylation of cytosines, histone modifications, chromatin remodelling enzymes and non-coding RNA molecules are important factors. In the theoretical section of this work, the epigenetic regulation of gene expression is summarized, with the focus on the methylation of cytosines. Most attention is devoted to the regulation of catalytic subunit of telomerase and to epigenetic modifications of telomeres.Experimental part of the thesis deals with the analysis of telomerase activity in tissues of A. thaliana, the transcription of the gene coding for the catalytic subunit of telomerase, and epigenetic modifications of chromatin of this gene in wild type plants and in plants with T-DNA insertions in DDM1 and MET1 genes. Loss of function of these genes leads to the reduction of the overall methylation in A. thaliana genome. Furthermore, the length of telomeres and methylation of telomeric cytosines are analyzed in plants with hypomethylated genome. In the section „Discussion“, the results of experiments are discussed in connection to the current state of knowledge of the epigenetic regulation of telomerase and telomeres in different model organisms.
