Abstract:

Předložená diplomová práce je zaměřena na studium retenčního chování trinukleotidů pomocí vysokoúčinné kapalinové chromatografie na obrácené fázi v iontově interakčním módu. Ke studiu retence těchto krátkých nukleotidů byly aplikovány pokročilé chemometrické techniky a to metoda plánování pokusů a umělé neuronové sítě. První část práce je věnována metodě plánování pokusů, kde slouží jakožto optimalizační nástroj pro zvýšení kvality separace látek a prostředku pro studium separačního systému. Spolu ve spojení s umělými neuronovými sítěmi je dále testována schopnost predikce retenčního času neznámé sekvence či její identifikace ve směsi. V neposlední řadě byla blíže zkoumána sekvenčně retenční závislost, která byla již zjištěna v rámci předchozích studií.

Abstract:

This diploma thesis is focused on study of retention behaviour of trinucleotides by high performance liquid chromatography. Separation was based on reversed phase chromatography in ion interaction mode. Advanced chemometric techniques such as experimental design and artificial neural networks were applied to study the retention of these short nucleotides. The first part of thesis is devoted to the experimental design method, where it serves as an optimisation tool for improving of separation quality and a mean for the study of the separation system. Together, in conjunction with artificial neural networks, an ability to predict the retention time of unknown sequence and its identification in the mixture is tested. Last but not least, the sequence-dependent retention which has been observed in previous studies was deeply investigated.
