SREBP1 gene variability and association with selected meat quality traits of cattle

Abstract:

Genetické poznatky přinášejí velké výhody pro dnešní živočišnou výrobu, kde pomáhají zrychlit produkci, selekci a genetický zisk pro dané vlastnosti. Předkládaná diplomová práce zkoumá gen SREBP1 na populaci českého strakatého skotu. Zmíněný gen reguluje především lipogenezi, dále je spojován se složením mastných kyselin v intramuskulárním tuku a zároveň ovlivňuje kvalitu masa. Intramuskulární tuk velmi významně působí na senzorické vlastnosti masa, především na chuť, šťavnatost a křehkost. Tato práce byla zaměřena na stanovení genotypů u cca 100 kusů býků plemene ČESTR. Bylo stanoveno celkem 95 genotypů genu SREBP1 pomocí amplifikace PCR a vizualizace byla provedena prostřednictvím horizontální agarózové gelové elektroforézy, následně byly některé vzorky ověřeny metodou Sangerova sekvenování. U vybrané populace byly určeny pouze dva typy genotypů: LL a LS, genotyp SS se v této populaci neobjevil. V dané populaci se nejvíce vyskytovali homozygoti LL (n=71) a dále heterozygoti LS (n=24). Frekvence genotypů byla následovná: LL 74,7 % a LS 25,3 %. Relativní frekvence byla stanovena u alely L na 87,35 % a u alely S na 12,65 %. Dále byla stanovena Hardy-Weinbergova rovnováha s výsledkem, že zkoumaná populace je v genetické rovnováze. Mezi genotypy a IMT nebyla identifikována asociace, takže IMT průkazně nevyšel (p=0,6974). Ovšem statisticky významný rozdíl (p=0,0076) byl prokázán u barvy.

Abstract:

Genetic knowledge brings great benefits to today's animal production, where they help speed up production, selection and genetic gain for given traits. In this work, the gene SREBP1 is examined on the population of Czech Fleckvieh cattle. This gene regulates mainly lipogenesis, however it is also associated with the composition of fatty acids in intramuscular fat, as well as it affects the quality of meat. Intramuscular fat has a very significant effect on the sensory properties of meat, especially on taste, juiciness and tenderness. This work was focused on determining the genotypes of 100 bulls of the Czech Fleckvieh cattle breed. A total of 95 genotypes of the SREBP1 gene (71x LL, 24x LS, 0xSS) were determined by PCR amplification and visualization was performed by horizontal agarose gel electrophoresis. Some samples were then verified by sanger sequencing. After that the genes were associated with selected meat quality traits. In this population of randomly selected bulls, the Hardy-Weinberger equilibrium was determined and an association analysis was performed. Only two types of genotypes were determined in the selected population: LL and LS, the SS genotype did not appear in this population. LL homozygotes (n = 71) and LS heterozygotes (n = 24) were the most common in the given population. The frequency of genotypes was as follows: LL 74.7 % and LS 25.3 %. The relative frequency was defined to be 87.35 % for the L allele and 12.65 % for the S allele. Furthermore, the Hardy-Weinberg equilibrium was specified, resulting that the studied population is in genetic equilibrium. No association was identified between genotypes and IMT, so IMT did not appear conclusively (p = 0.6974). However, a statistically significant difference (p = 0.0076) was demonstrated in color.
