Functional Connectivity-Based Biomarkers for Brain Disorders

Abstract:

Tato diplomová práce se zaměřuje na identifikaci biomarkerů onemocnění mozku u pacientů s hraniční poruchou osobnosti. Jejím hlavním cílem je pomocí metod funkční konektivity tyto biomarkery najít. K tomuto účelu se využívá dat z funkční magnetické rezonance, která doká-žou zachytit aktivní oblasti v mozku při vykonávání určitého kognitivního úkolu. Předložená diplomová práce zahrnuje kromě základních informací o tomto duševním onemocnění také rešerši literatury z oblasti anatomické, funkční i efektivní konektivity, a dále popisuje metody pro konektivitu funkční. V praktické části jsou teoretické poznatky aplikovány na datový sou-bor ze studie Impulzivita, který zahrnuje 72 osob – 36 zdravých dobrovolníků a 36 pacientů s hraniční poruchou osobnosti. Nejdříve jsou popsány základní charakteristiky datového sou-boru, poté jsou pomocí vývojového prostředí Matlab a toolboxů SPM 12 a CONN obrazová data předzpracována, transformována do MNI stereotaktického prostoru, a nakonec jsou na ně aplikovány metody funkční konektivity pro identifikaci biomarkerů. V závěru práce jsou do-sažené výsledky diskutovány.

Abstract:

This diploma thesis focuses on the identification of biomarkers in brain images of pa-tients with a borderline personality disorder. Its main goal is to find these biomarkers using functional connectivity engines. For this purpose, functional magnetic resonance imaging da-ta is used, which can capture active areas in the brain while performing a certain cognitive task. The presented diploma thesis includes, in addition to basic information about this mental illness, also a search of the literature in the field of anatomical, functional and effective con-nectivity, and further describes methods for functional connectivity. In the practical part, the theoretical knowledge is applied to the data set from the study Impulsivity, which includes 72 people - 36 healthy volunteers and 36 patients with a borderline personality disorder. First, the basic characteristics of the data set are described, then using the Matlab development envi-ronment and the SPM 12 and CONN toolboxes, the image data are preprocessed, transformed into an MNI stereotactic space, and finally, functional connectivity methods are applied to identify biomarkers. At the end of the work, the achieved results are discussed.
